In this paper, the experiment chooses the fruit from Qaidam Basin in the Qinghai-Tibet Plateau as the research material to study the trace element content and obtain polysaccharide by microwave-assisted extraction and ultrasound extraction. Explored the solid to liquid ratio, extraction temperature, extraction time and the number of single-factor experiment and orthogonal experiment, Investigated the optimum technological condition of that. And various pharmacological activity of Nitraria tangutorum Bobr fruit polysaccharides.1.The content of 8 trace elements K、Ca、Na、Mg、Cu、Zn、Fe and Mn 8 kinds of microelement and Pb、As、Hg 3 kinds of heavy metal elements in Nitraria tangutorum Bobr fruit pharmacological from Qaidam Basin in the Qinghai-Tibet Plateau was determined by atomicabsorption spectroscopy. Experimental results show that the content of Na, K, Ca is more than other berries. The content of heavy metal Pb, As and Hg are within the scope of the food safety standards set by the state, in line with the national food safety regulations.2.Assisted by microwave-assisted extraction and ultrasound extraction to extract polysaccharide from Nitraria tangutorum Bobr fruit.Determined the optimum technological condition was: Microwave power of 1000 w, extract time of 5 minute, supersonic extraction condition solid-liquid ratio is 1:5, extraction temperature is 95℃, extraction time is 1 h, and the number of ultrasound extraction is 3 times.Through the proof test, the average Nitraria tangutorum Bobr polysaccharides content is 33.875 mg·g-1.The yield of Nitraria tangutorum Bobr polysaccharides is stable and the test is reliable.3.Research on the antioxidant effect in Nitraria tangutorum Bobr fruit polysaccharide.With the Nitraria tangutorum Bobr polysaccharides for the reagent, through determining the biochemical indicators(SOD in mice serum superoxide dismutase, The vitality of serum malondialdehyde MDA) related aging metabolic enzymes after free swimming of mice to study the antioxidant effect in Nitraria tangutorum Bobr polysaccharide. Experimental results show that Nitraria tangutorum Bobr polysaccharide can improve the vitality of serum SOD and the vitality of serum MDA. Therefore, good antioxidant function was discovered in Nitraria tangutorum Bobr polysaccharide.4.Research on the anti-fatigue effect in Nitraria tangutorum Bobr fruit polysaccharide.With the Nitraria tangutorum Bobr polysaccharides for the reagent, through the weight loading swimming experiment, free swimming experiment and fatigue related biochemical indicators(LD, BUN, liver glycogen) of mice to study the anti-fatigue effect in Nitraria tangutorum Bobr polysaccharide. Experimental results show that Nitraria tangutorum Bobr polysaccharide can significantly extend the exhaustion of mice swimming time(P<0.01) in weight loading swimming experiment. reduce the serum LD(P<0.01)and serum BUN concentration, increase liver glycogen(P<0.01)in free swimming experiment. Therefore, good anti-fatigue function was discovered in Nitraria tangutorum Bobr polysaccharide.5. Research on the anti-stress effect in Nitraria tangutorum Bobr polysaccharide.With the Nitraria tangutorum Bobr polysaccharides for the reagent, through resistant to high temperature of 45℃, resistant to low temperature of-5℃ and anti-hypoxic experiment(atmospheric oxygen) to study the anti-stress function in Nitraria tangutorum Bobr polysaccharides. The experimental results show that, in resistance to high temperature experiment, Nitraria tangutorum Bobr polysaccharides can extend the survival time of mice in high temperature environment, the high dose group can extend the time of survival 10.8 min,the low dose group can extend the time of survival 6.1 min. In resistance to low temperature experiments, Nitraria tangutorum Bobr polysaccharides can extend the survival time of mice in low temperature environment, the high dose group can extend the time of survival 14.7 min,the low dose group can extend the time of survival 13.1 min.In anti-hypoxic experiment, the high dose group can extend the time of survival 10.1 min,the low dose group can extend the time of survival 6.9min.. The result indicated that the anti-stress function of Nitraria tangutorum Bobr polysaccharide was not significant.6. protective effect of liver injury induced by chemical reagent in Nitraria tangutorum Bobr polysaccharide.With the Nitraria tangutorum Bobr polysaccharides for the reagent, The mice were randomly divided into four experimental groups according to their weight, high dose groups and low dose groups, the normal control group, comparison groups of Bifendate Tablets, After administration of 15 days, their liver were injured by CCl4. determining theglutamic-pyruvic transaminase enzyme(ALT) and glutamic-pyruvic transaminase(AST) in mice serum. The results show that Nitraria tangutorum Bobr polysaccharide could obviously control the content of ALT, AST in serum.The result indicated that Nitraria tangutorum Bobr polysaccharide has significant protective effect on liver injury induced by chemical reagent.
