Objective To explore the treatment effect and its possible mechanism of salvianolate in vascular endothelial dependent vasodilation dysfunction of carotid atherosclerosis rats; To investigate the correlation between degree of carotid atherosclerosis and serum CyPA 、 8-iso-PGF2α concentration in Patients with carotid atherosclerosis.Methods 1.Forty male Wistar rats were randomly divided into the control group, the CAS model group, the high dose and low dose of salvianolate treatment groups, ten in each group. The CAS model group and salvianolate treatment groups were given intraperitoneal injection a single dose of Vitamin D3 with high cholesterol diet, after 15 weeks, the salvianolate treatment groups were given intraperitoneal injection with a high dose of salvianolate with 30mg/kg.d, and a low dose of 10mg/kg.d for 3 weeks. The control group were fed with base diet with thesame dose of saline injection. The carotid artery were separated from executed rats. The 3mm vessel rings’ endothelial dependent vasodilation were observed. Other parts were stained with hematoxylin and eosin, determin the ratio of calcified plaque area in the section area. And immunohistochemical staining detected the expression of CyPA、KLF2、eNOS.2.Selecte 85 cases of patients with acute cerebral infarction, confirmed carotid atherosclerotic plaques by carotid ultrasound.Divided into single、multiple carotid plaques groups. Selecte 24 cases of patients without acute cerebral infarction and carotid atherosclerotic as control group. The serum Cy PA 、8-iso-PGF2α concentration in each group were measured by ELISA.Compare atherosclerosis risk factors of each group and compare difference of serum CyPA 、 8-iso-PGF2α concentration.Analyze the relationship between serum CyPA 、8-iso-PGF2α concentration and degree of carotid atherosclerosis.Results 1.The vasodilatation of low dose salvianolate group is close to the CAS model group(P>0.05). Vasodilatation of the high dose group improved compared with the CAS model group under the 10-6-10-4 mol/L acethylcholine(P<0.05),but the difference with the control still significant(P<0.05).HE staining shows calcified plaque in the two salvianolate groups and the CAS model group,the lesions area of them have no difference( P > 0.05). The mean light density analyze shows decrease CyPA expression decrease, KLF2、eNOS expression increase compared with the CAS model group( P < 0.05). These three indexes’ expression have no difference with the CAS model group(P>0.05).2.Comparison of three groups patients with basic data shows that carotid plaques groups of smoking, and hypertension, coronary heart disease history, TIA or stroke history, triglycerides, diabetes proportion is higher than that in control group(P>0.05).The diabetes, FPG of multiple carotid plaques group,is higher than single carotid plaques group and the control group.Serum high density lipoprotein in Single and multiple carotid plaques groups is higher than controls(P<0.05).Serum CyPA, 8- iso- PGF2 alpha of multiple carotid plaques group increased significantly, the difference was statistically significant between the three groups.Serum CyPA, 8- iso- PGF2 alphalinear correlation.Conclusion 1. Salvianolate can improve the vasodilation dysfunction of CAS rats, may relate to the CyPA/KLF2/eNOS signal transduction pathway.2.Oxidative stress is involved in the formation of carotid plaques, Serum CyPA, 8- iso- PGF2 alpha may be relate to number of carotid plaques.CyPA can be a sensitive index of the oxidative stress levels in the body.
