Purpose:To screen the high-expression genes in patients with high squamous intraepithelial lesion (HSIL) with Oncomine database, we compared the expressions of these genes in HSIL patients by using real-time fluorescent quantitativePCR (qPCR). We aimed to determine the abnormal expressing genes, and their correlation with early-stage cervical cancer.Methods:High-expression genes in HSIL were explored with Oncomine database, among which,8genes were selected for further study after weighting analysis.12cases of HSIL patients were enrolled and treated with cervical conization. The lesion tissues and normal cervical tissues were then selected from the section slides of cervical tissues for extracting their RNA and reversely transcripting into cDNA. QPCR technology was then used for testing. Difference expression of each gene was then calculated by following equation2△△Ct=2△Ct Lesion tissue/2△Ct Normal tissue. And the expressions of the upregulated genes in HSIL patients were further compared with paired-t test.Results:Aight genes with high expression level were selected from the HSIL patients using Oncomine database, and7genes with high expressions in HSIL were confirmed through qPCR detection and data statistical analysis. They were MMP1, SPP1, APOC1, MMP12, ECT2, IDO1and HELLS. All genes were found to be highly expressed in patients with HSIL, presenting statistically significant difference (P<0.05). Only one gene (NEK2) with its expression had no statistically significant difference in this experiment.Conclusions:The Oncomine, based in large sample data, provides advantage for digging the HSIL-related genes in an easy and accurate manner. Herein,7genes with high co-expression levels in cervical squamous-cell carcinoma (CSCC) and HSIL may be involved in the process of HSIL’s progressing into CSCC, which expands the early warning indicator system of CSCC, and offers scientific basis for early detection, prevention and effective treatment of cervical cancer, and lays the foundations for the further studies in future.
