Part one:The clinical research about changes of ACE2-ACE and inflammation in diabetic mellitus.Objective:To explore the role of ACE2-ACE balance and inflammation in diabetic mellitus,we detected the level of serum ACE2,ACE,MCP-1,VCAM-1 in patients with diabetes mellitus and healthy person..Methods : 38 diabetic patients without chronic complications were choosed as simple DM group and 35 well-beings as the control. Basic informations of subjects including age, duration of DM,gender were recorded. Venous blood samples were gathered for estimating fasting blood glucose(FBG),Hb Alc and serum lipid profile. Serum ACE2 、ACE、MCP-1 and VCAM-1 were measured by using ELISA kits of all samples.Results: The serum level of ACE2 in simple DM group(16.78 ±0.87 ng/ml) was higher than that in control group(13±1.41 ng/ml,P <0.05). The serum level of ACE in simple DM group(26.36 ±2.10 ng/ml)was higher than that in control group(18.51±2.51 ng/ml,P <0.05). The serum level of MCP-1 in simple DM group(37.7 ±4.09 pg/ml)was higher than that and control group(22.63±4.04pg/ml,P<0.05)。 The serum level of VCAM-1 in simple DM group(257.04 ±19.48 ng /ml)was higher than that in control group(221.81±12.06ng/ml,P <0.05)Conclusion:The ACE2- ACE activation and inflammatory cytokines: VCAM,MCP –1 are essential in the development of diabetic mellitus.Part two : The effects of ACE2 overexpression on inflammatory response in experimental diabetic cardiomyopathyObjective: To observe the impact of ACE2 gene treatment on inflammation and oxidative stress in DCM.Methods :DCM was induced by intraperitoneal injection of STZ in Wistar rats. Diabetes was induced by intraperitoneal injection of STZ in rats. After 12 weeks, the rats were randomly divided into three groups: the ACE2 group, the EGFP group and the control group. After gene transfection,the protein expression of ACE2、MCP-1 、 VCAM-1 、 IL-8 、 macrophage and 4-HNE were analyzed by immunochemistry.The protein expression of P22,ACE and NF-KB were analyzed by western blot.Results.The protein expression of NF-KB、MCP-1、VCAM-1、IL-8,macrophage and 4-HNE were significantly lower in ACE2 gene treatment group than that in DCM and EGFP groups..Conclusion ACE2 inhibit inflammation and oxidative stress in DCM.
