Objective Analysis of cyclooxygenase-2(COX-2) and carcinoembryonic antigen(CEA) expressio n in different stages of development of colorectal cancer. Study the role of the relationsh ip between COX-2 and CEA in colorectal tumorigenesis and various clinicopathological features, and both expressed in colorectal adenocarcinoma correlation.Methods Randomly selected from Dalian Central Hospital Pathology Department in July 2012-July 2014 after surgical resected colorectal adenocarcinoma archived paraffin slices 50 cases, while selecting the distal colorectal adenocarcinoma pathologically confirmed 50 cases of normal colorectal mucosa tissues, digestive endoscopic biopsy specimens of colorectal adenomas in 50 cases, Using the immunohistochemical staining(SP), three specimens of expression of COX-2 and CEA were detected. The results of immunohistochemical staining criteria. The results of immunohistochemical staining criteria: protein localization in cytoplasm stained brown cells, counting 10 high power field(HPF)staining positive cells accounts for a percentage of the total.Positive staining cells were 0-10% as "-", 10-25% as "+" 25-50% as "++", 50-100% as "+++." Chi-square test is used to analyze cox-2 and CEA between colorectal adenocarcinoma, adenoma and normal colorectal mucosa of expression and clinicopathologic feature of patients. Application Spearman correlation analysis and correlation of COX-2 and CEA expressed in colorectal tumors.Results 1. COX-2 positive staining in cytoplasm and cell membrane, stained brown cells. The degree of expression of COX-2 was categorized into 4 grades ranged from “-” to “+++”,the expression grades in series in normal colonic mucosal tissues was negative,respectively; that in adenomas was 52.0%,28.0%,16.0%,4.0%, respectively, that in adenocarcinoma was 20.0%,20.0%,30.0%,30.0%, respectively. Colorectal cancer, colorectal adenomas COX-2 expression rates were significantly higher than in normal mucosa(P <0.05), there was a significant difference in comparison(P <0.05) colorectal adenocarcinoma and colorectal adenomas. 2. COX-2 expression and clinicopathologic features of colorectal cancer relationship: COX-2 protein expression and colorectal adenocarcinoma differentiation relations, from well-differentiated to poorly differentiated was 42.8%,87.5%,100.0%; respectively. COX-2 expression level was significantly correlated with carcinoma tissue cell differentiation degree(P<0.05), from high to medium to poorly differentiated colorectal adenocarcinoma protein expression between differentiation of large intestine cancer was on the rise. Colorectal adenocarcinoma COX-2 expression in lymph node metastasis was significantly higher than that without lymph node metastasis,(P <0.05), and were positively correlated, and adenocarcinoma C + D stage was significantly higher than A + B.COX-2 expression had no correlation with patients’ age, sex, location.3. CEA expression and clinicopathologic features of colorectal cancer relationship:CEA protein expression and colorectal adenocarcinoma differentiation relations, from well-differentiated to poorly differentiated was 35.7%,81.3%,100%; respectively. CEA expression level was significantly correlated with carcinoma tissue cell differentiation degree(P<0.05), from high to medium to poorly differentiated colorectal adenocarcinoma protein expression between differentiation of large intestine cancer was on the rise. Colorectal adenocarcinoma CEA expression in lymph node metastasis was significantly higher than that without lymph node metastasis(P <0.05), and were positively correlated, and adenocarcinoma C + D stage was significantly higher than A + B.CEA expression had no correlation with patients’ age, sex, location.5. Between COX-2 and CEA in adenocarcinoma tissue protein expression level was statistically significant(P<0.05) were positively correlated.Conclusion: 1.The positive expression of COX-2 in normal colorectal mucosa, adenoma, colorectal cancer on the rise. 2. The positive expression of CEA in normal colorectal mucosa,adenoma,colorectal cancer on the rise. 3.COX-2 expression is significantly correlated with lymph node metastasis and Dukes stage in colorectal adenocarcinoma. COX-2 expression has no correlation with patients’ age, sex, location.4.CEA expression is significantly correlated with lymph node metastasis and Dukes stage in colorectal adenocarcinoma. CEA express ion has no correlation with patients’ age, sex, location. 5. COX-2 expression was positively correlated with CEA in colorectal ade- nocarcinoma.
