Objective:This paper is to discusses the influence of the proportion of PLGA and pore-foaming agent NaCl on the scaffold porosity through preparing the PLGA porous scaffold with solvent casting particle leaching method, and study the adhesion effeciency of the marrow mesenchymal stem cell on the scaffold after its surface is modified with gelatin. Methods: 1.Apply solvent casting particle leaching method to prepare the scaffold in the proportion of 8/1, 9/1, 10/1, 9/1, 12/1 of PLGA/Na Cl, record the influence of each proportion has left on the scaffold porosity through measurement, and modify the surface of 3D-scaffold with 0.2% gelatin solvent. 2.Extract and culture the bone marrow mesenchymal stem cells of an adult rat, implant the third generation of the cells after the extraction on the scaffold,culture them for 5 days and observe the growth states respectively on modified and non-modified surfaces by scanning with electron microscope. Results:1. The prepared scaffold is porous and well-connected in pores under the observation of electron microscope; 2. The porosity of the scaffold which is prepared in the proportion of 8/1, 9/1, 10/1, 9/1, 12/1 of PLGA/NaCl are 83.1,85.8,87.5,91.7,92.1 respectively; 3.The bone marrow mesenchymal stem cells grow better on the surface-modified scaffold than that on the non-modified scaffold. Conclusion:1. Scaffold with high porosity and pore connectivity can be prepared by solvent casting particle leaching method; 2.As the proportion of PLGA/NaCl increases, the porosity of the PLGA scaffold will rise; 3.The PLGA scaffold which is modifed by gelatin on its surface has better cellular affinity and higher adhesion rate than the non-modified one.
