| Tea polysaccharides are a widespread natural plant polysaccharides extracted from tea. The extensive experimental research shows that tea polysaccharides have various functions such asimmuno-stimulating activities, anti-hyperglycemic, anti-aging, anti-tumor activities, immune enhancement, anti-diabetes. The structure and biological activity of tea polysaccharides have differences in varieties and regions. Wuyuan green tea, as Chinese tea representative, is very famous for its good quality, high productivity and convenient material source.This investigation focuses on the biological activity of tea polysaccharides. Polysaccharides were isolated by water extraction, alcohol precipitation, deproteinization and freeze-drying. Its final purity is over 95%. On the basis of existing research, the technology of modern immunology and molecular biology can be used to study the regulation effects of tea polysaccharides shown on immune cells and to explore its possible immunoregulatory mechanisms, which could be of great theoretical significance and clinical practical value for understanding the immune pharmacological effect of tea polysaccharides.To assess the impact of tea polysaccharides immune function in vitro, immunosuppressed mice macrophages were used as cell model. Research results showed that it can significantly enhanced the phagocytosis of peritoneal macrophages, increased the secretion of reactive oxygen species, and improved the cytokine secretion level of TNF-α, IL- 6, IL- 1β. RT-PCR and Western blot results showed that tea polysaccharides can improve the abdominal macrophage iNOS, TNF-α, IL-1β mRNA expression level and increase the expression of iNOS protein. So it is shown that tea polysaccharides can enhance the immunosuppressed mice abdominal macrophage function.Immunosuppressed mice spleen lymphocytes were used as cell model to assess biological activity of tea polysaccharides. The results showed that tea polysaccharides can significantly promote the immunosuppressive mice spleen lymphocyte proliferation and also has synergistic effect with ConA. Tea polysaccharides increased the percentage of T lymphocytes CD3+, CD4+, raise the ratio of CD4+/ CD8+, thus against immunosuppression caused by cyclophosphamide. It can promote the IL-2 and IL-4 secretion and the secretion of NO. From the above, it suggests that tea polysaccharides have immune function on immunosuppressed mice spleen lymphocytes.The above studies show that tea polysaccharides have biological activity of regulating body immunity, especially for the two major immune cells- abdominal macrophage and spleen lymphocyte.However,the immune regulation mechanism still needs further research. |
