[Objective]:We study the effect of Allicin Injection treatment on cell proliferation, apoptosis, migration and invasion on human cholangiocarcinoma cell line QBC939.[Methods]:We performde MTT assay to measure cell viability of human cholangiocarcinoma cells treated with different Allicin concentrations at different time intervals, we determined the optimum Allicin treatment concentration and time point and calculated the IC50 according to cell growth curve. We also recorded the cell cycle and cell morphological changes under microscope. We used flow cytometry to analyze cell cycle and cell apoptosis of the Allicin treated cells. At last, we performed cell scratch test to investigate cell migration and invasion abilities.[Results]:More than 95% human cholangiocarcinoma cells are alive in-vitro culturing. The cells reached the logarithmic grow phase after 24 hours cell passage. The MTT assay showed time dose dependent effect of the Allicin and the calculate IC50 is 7.25mg/L. Flow cytometry analysis showed on going cell apptosis and cell cycle arresting at G2/S period. The cell scratch test demonstrated that the Allicin prohibited cell migration and invasion.[Conclusion]:Allicin prohibits human cholangiocarcinoma cell proliferation in a does and time dependent manner. Allicin induces cell apoptosis and arrests cell cycle at G2/S period. Allicin also prohibits cell migration and invasion. Therefore, we postulate that Allicin treatment has potentials in treating hunman cholangiocarcinoma.
