Objective:1.To determinate the levels of microRNA-27a expression in hepatocellular carcinoma; 2.To observe the effect of microRNA-27a on the formation of hepatocellular carcinoma.Method:1.We find that the levels of miR-27a expression is abnormal by data access,but there is no study about the expression in hepatocellular carcinoma;2. Culturing hepatocellular cell line HEPG2 and normal liver cell by cell culture technology;3. Determinating the levels of microRNA-27a expression in cells by RT-PCR technology and representing Ct.4.To compare two sample for means if there is a difference by t-test,and to obtain the expression of target gene in hepatocellular carcinoma is higher or lower.5.We collected tissue samples from patients with HCC and serum AFP, then research the relationship between the levels of microRNA-27a expression and serum AFP throughX2 test.Result:1.There are expressions in normal liver cells and hepatocellular carcinoma cells.2.Gel electrophoresis show that we get the target gene by PCR amplification.3.The △Ct values of microRNA-27a in hepatocellular cell line HEPG2 is 8.8±0.6 and27.6 ±0.6 in normal liver cells.4. The △Ct values of miR-27a in hepatocellular cell line HEPG2 and in normal liver cells are different. The △Ct values of miR-27a in hepatocellular cell line HEPG2 is higher than in normal liver cells (t=132.1,p<0.001).5.There is no difference between serum AFP and △Ct values of miR-27a in patients with hepatocellular carcinoma (X2=1.3, P=0.25)Conclusion:1.There is difference between hepatocellular cell line HEPG2 and normal liver cells about the expression of miR-27a;2.The expression of miR-27a in hepatocellular cell line HEPG2 is higher than in normal liver cells,suggesting this gene may play a role of carcinoma genes in the development of hepatocellular carcinoma.3.There is no difference between serum AFP and △Ct values of miR-27a in patients with hepatocellular carcinoma.
