Objective:To observe the effect of acupuncture therapy on the role in regulation of calcium and the influence of different treatment time on curative effect, by means of determining the substantia nigra Ca2+ content changes in the brain of rats at different time points (14d,28d) by Fura-2 fluorescence probe method.Methods:First of all, build a model of rat which suffers from Parkinson’s disease. Then choose two target points to inject 6-hydroxydopamine (6-OHDA) in one side of the rat’s striatum. We can use the rotational behavior caused by injection of apomorphine (apo) to judge whether the experiment is successful. The Groups are as follows:music electro acupuncture group 12 rats, model group 12 rats and sham operation group 12 rats, control group 12, Madopar in the control group (n= 12) and pulse EA group (n= 12). Each group was divided into14d and 28d group according to the treatment time. Treatment methods:In the music electro acupuncture group, insert the needle from Point Baihui to the temple. After the needle penetration, connect music electro acupuncture. The control group is to give rats intragastric administration of Madopar. After successfully molding, anesthetize the rats on the 14th day and 28th day respectively in each group of the treatment. And then use 4% poly formaldehyde brain irrigation ECM to make it fix and isolate the substantia nigra. Use Fura-2 fluorescence probe method to determine the content of substantia nigra Ca2+ in rat’s brain. Statistical software SPSS 18.0 is used in data analysis in this research.Results:After molding, the14-day and 28-day Ca+ model group was significantly higher than the control group and sham operation group (P<0.01); 14 days later, in the music electro acupuncture treatment group and pulse electro acupuncture treatment group, Ca2+ contents were lower, but there was no significant difference between the two groups (P>0.05). But on the 28th day,, there was significant difference between music electro acupuncture treatment group and pulse electro acupuncture group (P<0.05); in all time point of Madopar and music electro acupuncture group the content of Ca2+ is low, and there was no significant difference between the two groups (P>0.05); in the Madopar and pulse electro acupuncture therapy group, there was significant difference (P<0.05); no significant difference was found in the control group and the sham operation group at all time points (P>0.05).Conclusion:Both Music electro acupuncture treatment and pulse electro acupuncture treatment can reduce the increased content of substantia nigra DA neurons Ca2+ caused by PD. When compared with the music electric acupuncture, the pulse electro acupuncture has better tolerability but less healing effect.
