Objective: Retrospective study of patients with lower limb deep venous thrombus,according to long-term blood coagulation routine monitoring results to adjustment warfarin dosage to the actual amount of, and the correlation between rapid gene detection calculated theory dosage. Because gene detection can accurately to provide experience and basis for the recommended dosage of warfarin.Methods: Retrospective analysis of vascular surgery, the First Affiliated Hospital of Wan nan Medical College during 2013.01-2015.01 in hospitalized patients with lower extremity deep vein thrombosis in 31 patients simple deep venous thrombosis(except hypertension, diabetes and other basic diseases to minimize the drugs to warfarin metabolic effects). On admission, peripheral venous blood samples 2 ml for CYP2C9 and VKORC1 Gene test, and then according to the test results, combined with patient’s age, gender, height, weight, According to the model calculation formula of warfarin IWPC recommended by international warfarin, drug genome Federation,calculated the theoretical dosage and register. But the patient is not in gene detection calculated theoretical dosage administration, and to the coagulation regular monitoring of blood conventional constantly adjust the dosage of warfarin, until the patients( INR) maintained at 2-3, registration at the actual dosage. Retrospective analysis of relevance at the actual dosage and early gene detection theory the recommended amount, understand CYP2C9 and VKORC1 gene test on patients with lower limb deep venous thrombosis warfarin dosage has the function of guidance and its advantages and disadvantages.Results: Retrospective analysis found: traditional by day periodic monitoring of clotting routine actual dosage adjustment, and have correlation with the theoretical recommended amount by fast CYP2C9 and VKORC1 Gene detection calculate, up to 40%.Conclusion:1、 To deep venous thrombosis patients, the theoretical recommended amount by fast CYP2C9 and VKORC1 Gene detection calculate. have correlation with traditional periodic monitoring of clotting routine actual dosage adjustment. CYP2C9 and VKORC1 Gene detection can be used to guide clinical drug warfarin.2、CYP2C9 and VKORC1 Gene detection, the results can be quickly, quickly calculate the theory the recommended amount,saving time, effort. The monitoring of regular blood coagulation adjustment actual dosage need more time,ranging from 3-30 days, time-consuming and laborious.In short, CYP2C9 and VKORC1 Gene detection, which can effectively and quickly provide guidance dosage of warfarin for lower limb deep vein thrombosis patients.
