Objective Through establishing SD rat model of insulin resistance, morphological changes were observed in skeletal muscle tissue and measuring the change in the expression of GLUT 4, to discuss the effect of resistance training exercise in experimental observation of GLUT 4 in rat model of insulin resistance mechanisms.Methods 28 SD male rats after one week of adaptation process, were randomly divided into four groups, namely group SD1, group SD2, group SD3, group SD4, group SD1, SD2, SD3, SD4, also have 7 rats each group. Group SD1 is control group, SD2 is model DM group, SD3 is exercise group, SD4 is resistance training group. Group SD1 and SD2 did not receive any treatment, the rest of group SD2, SD3, SD4 were established for rat model of insulin resistance (High fat diet feeding method to establish the mechanism of insulin resistance in rats). Group SD3, SD4 do training five days a week, for four weeks. Group SD3 for first two weeks do 40 minutes exercise everyday, two weeks later do exercise 60 minutes everyday. Group SD4 do exercise 30 minutes everyday. All were killed after four weeks of training. Each group of rats were observed for generally pathology and morphology, detection of glucose transporter 4 (GLUT4) expression.Results Four weeks after the intervention compared with before intervention, blood glucose level of resistance training group show a significant decrease (P<0.05), exercise group did not show significant differences, blood glucose level of model DM group show significant increase (P<0.05), control group did not show significant differences; four weeks after intervention GLUT 4 level of model DM group and control group did not show significant differences, GLUT 4 level of resistance training group and exercise group is higher compare to model DM group, but resistance training group show more significant differences (P<0.01); at the same time, morphological observation show that skeletal muscle injury level of resistance training group and exercise group is less than model DM group.Conclusion Resistance training may play a important role in the intervention of GLUT 4 expression in rat model of insulin resistance, by lowering blood sugar, increased expression of GLUT4, a rat model of insulin resistance show a significant improvement.
