ObjectiveIn this research, LPS-induced pulp pain in rats,real-time quantitative polymerase chain reaction technique was used to observe the effect of EA ‘Hegu’ on the expression of trigeminal ganglia P2 X receptor m RNA, further research P2 X receptor in the information transport process,in order to explore the mechanism of EA " Hegu " treatment.Methods42 SD rats were randomly divided into six groups, N group, C group, M group, A group, E group, AE Group.N group: no processing; C group: the pulp cavity drilled and injected with the same dose of pulp pain saline infiltration,fillings will be closed cavities 5-6 min later; M group: both sides of the maxillary second molar drilling depth and exposed the pulp cavity with a small hand drill(drill diameter about 1mm), using a micro syringe concentration within 5ug/ul of LPS solution(per well injection of about 0.5ul/100g), and closed with a dental filling cavities infiltrating 5-6 min later; A group: modeling method was same as M group, the A-317491(0.5mg/kg) was injected into the pulp with LPS; E group:modeling method was same as M group, acupuncture treatment was started after induced method immediately,bilateral‘Hegu’were acupuncture with 1-2mm,30 min,once per day,three times in total; AE group: modeling method was same as A group, acupuncture treatment was started after induced method immediately, bilateral‘Hegu’were acupuncture with 1-2mm,30 min,once per day,three times in total;. All the rats were executed at three day,observe the changes of rat body weight and behavior, using rt-pcr technique to detect the rat dental pulp, trigeminal ganglion P2 X receptor m RNA expression.Result1. Body weight Changes in rats : body weight of rats in N group were stable growth; steady growth in body weight of rats in C group, but the weight gain of rats in group C group and N group increase slightly compared with a slow and statistically significant(p <0.01); M group, A group, E group weight gain and the AE group N group and C group compared to grow more slowly, and has a statistically significant(p<0.01); E group, AE group weight gain compared with the pulp pain grew rapidly, and has a statistically significant(p<0.01); A group weight slightly faster growth compared with the M group and statistically significant(p<0.05); AE group weight gain and antagonist group and EA group compared to grow faster, and has a statistically significant(p<0.01); E group weight gain compared with A rapid growth agent.2. Behavior Performance in rats : N group rats shiny fur, move freely; M group, A group, E group, AE group behavioral performance and C group and N group compared to active, and has a statistically significant(p<0.01); A group, E group, AE group behavioralperformance are quiet compared with the model group has a statistically significant(p<0.01).3. The expression of P2X2 receptor m RNA in each group of rats. The expression of P2X2 receptors in rat dental pulp m RNA in each group : rats in each group pulp P2X2 receptor m RNA expression; no significant difference(p>0.05) between group expression N group and C group P2X2 receptor m RNA; significantly increased P2X2 receptor m RNA M group than in group N group and C group(p<0.01); A group, E group, P2X2 receptor m RNA AE group were increased(p <0.05) compared with group N and C group; A group, E group P2X2 receptor m RNA AE group compared with M group had declined(p<0.01).The expression of each group rat trigeminal ganglion P2X2 receptor m RNA : the trigeminal ganglion of rats in each group wereP2X2 receptor m RNA expression were no significant differenceexpression N group and C group P2X2 receptor m RNA(p>0.05); M groupwere significantly increased, A group, E group, AE group P2X2 receptor m RNA than in N group and C group(p<0.01); A group, E group AE group than in the M group P2X2 receptor m RNA were decreased(p<0.05); P2X2 receptor m RNA expression of E group than AE grouphigher(p<0.05).4. The expression of P2X3 receptor m RNA in each group of rats.The expression of P2X3 receptors in rat dental pulp m RNA in each group : rats in each group pulp P2X3 receptor m RNA expression; P2X3 receptor m RNA has no significant difference between group expression N group and C group(p>0.05); M group, A group and E group P2X3 receptor m RNA than N group and C group were significantly increased(p<0.01); AE group P2X3 receptor m RNA increased slightly compared with N group(p<0.01); A group, E group P2X3 receptor m RNAAE group than M group were significantly decreased(p<0.01); A group and AE group P2X3 receptor m RNA were significantly decreased than the E group(p<0.01).The expression of each group rat trigeminal ganglion P2X3 receptor m RNA : the trigeminal ganglion of rats in each group were P2X3 receptor m RNA expression; no significant difference between N group and C group P2X3 receptor m RNA expression(p>0.05); M group, A group, E group were significantly increased(p<0.01) AE group P2X3 receptor m RNA compared with N group, C group were increased(p <0.05); A group, E group P2X3 receptor m RNA compared with AE group were significantly decreased(p<0.01) compared with group M; P2X3 receptor m RNA AE group was significantly decreased(p<0.01).5. The expression of P2X4 receptor m RNA in each group of rats.The expression of P2X4 receptor in rat dental pulp m RNA in each group : the rats in each group pulp P2X4 receptor m RNA expression; between expression N group and C group P2X4 receptor m RNA has no significant difference(p>0.05);M group, A group, E group, AE group P2X4 receptor m RNA expression were significantly higher compared with N group and C group(p<0.01); E group and AE group P2X4 receptor m RNA than in A group were reduced(p<0.01).The expression of each group rat trigeminal ganglion P2X4 receptor m RNA : the trigeminal ganglion of rats in each group were P2X4 receptor m RNA expression; no significant difference P2X4 receptor m RNA expression between N group and C group(p>0.05); M group, A group, E group P2X4 receptor m RNA were significantly increased(p<0.01) AE group than in N group, E group and AE group P2X4 receptor m RNA compared with C group were increased(p<0.05).Conclusion1. P2X2、 P2X3、 P2X4 receptors are P2 X receptor subtype, which participate in the inflammatory pulp pain information transfer of in rats induced by LPS.2. Electroacupuncture at‘Hegu’(LI 4) effect on the inflammatory pulp pain rats weight and pain reaction.3. Electroacupuncture at‘Hegu’(LI 4) effect on the inflammatory pulp pain is related to P2 X receptor subtype P2X2、 P2X3、 P2X4 receptors.
