Objective To investigate the effect of Schistosoma japonicum soluble egg antigens(SEA) and Sept4_i1 on apoptosis of human hepatic stellate cell line LX-2 and to explore the associated potential mechanisms.Methods1. The effect of SEA on apoptosis and on the expression of apoptosis-associated molecules of LX-2 was assessed by caspase-3 activity assay kit and Western Blot. The mechanism by which SEA could induce apoptosis on LX-2 cells was detected by Western Blot and immunofluorescence analysis.2. The effect of Sept4_i1 on apoptosis of LX-2 was assessed by Western Blot and TUNEL. The mechanism by which Sept4_i1 could induce apoptosis on LX-2 was detected by Western Blot.Results1. SEA upregulated the expression of caspase-3, DR5 and p53, and downregulated the expression of p-Akt and α-SMA in LX-2. The apoptosis could be reversed by PPARβ/δ agonist, si-p53 and shDR5. Furhermore, the increased expression of DR5 and p53 induced by SEA could be reversed by PPARβ/δ agonist. All the results showed that SEA could upregulated the expression of caspase-3 by downregulating the expression of p-Akt and upregulating the expression of DR5 and p53.2. Sept4_i1-induced apoptosis on LX-2 cells could be observed by TUNEL staining, accompanied with the upregulation of caspase-3, PPARγ and the downregulation of α-SMA. Overexpession of Sept4_i1 could also induce the downregulation of p-Akt and Bcl-2 expression. However, the expression of DR5 and p53 was not modulated by Sept4_i1. All the results showed that Sept4_i1 could upregulated the expression of caspase-3 by inhibiting the expression of p-Akt and Bcl-2.Conclusions1. SEA could induce apoptosis of LX-2 through Akt/p53/DR5 pathway.2. The apoptosis of LX-2 could be induced by Sept4_i1 mainly through Akt/Bcl-2 signal pathway.
