Objective:To understand the resistance status about Children with mycoplasma pneumonia in Jilin city and investigate the mechanism of drug resistance, in order to provide a laboratory basis for the future development of clinical treatment and rational choice of antibiotics. Methods:In 2014, 1272 specimens were collected form 0-15 years old children with respiratory tract infection in Children’s Hospital of Jilin City, Mycoplasma was cultured and drug sensitivity test was done for rapid screening of Mycoplasma; resistance to macrolides culture fluid DNA extraction; while the design of primer PCR amplification of 16 SrRNA gene and 23 SrRNA gene, agarose gel electrophoresis, the recovery of DNA sequencing, sequencing results compared with a Mycoplasma standard(M129) 23 SrRNA gene recorded in GenBank. Observe the mutation of it has no resistance gene loci. Results:In 1272 cases of culture and rapid identification of respiratory tract infection in children with mycoplasma pneumonia and 449 were positive, the positive rate was 35.30%. Between different age groups after statistical treatment(χ2=15.95 P<0.05) had significant difference. 449 positive cases respectively to 9 kinds of antibiotics susceptibility test of Mycoplasma mycoplasma culture result, the most sensitive to 99.3%, sparfloxacin, gatifloxacin 98.7% levofloxacin 95.6%, these three kinds of quinolone drugs generally applies only to adult, so drug sensitivity rate in more than 95%. Secondly, sensitive is the acetyl spiramycin 95.6%, azithromycin 92.4%; Mycoplasma drug resistance rate is the highest clindamycin 12.9%, followed by erythromycin, roxithromycin, clarithromycin 7.6% 9.6% 10.2%. 17 strains of rapid cultured positive resistance to macrolides and extraction of DNA, 16 SrRNA and 23 SrRNAPCR were amplified, 17 strains have been obtained 16 SrRNA 277bp fragment and 23SrRNA792 bp fragment, the positive rate of both 100%; fragment sequencing of PCR amplified 23 SrRNA 792bp mesh results show, and the NCBI have been recorded MP standard strain M129 gene sequence alignment, 3 strains had 2063 sites of A G mutation, mutation and there were no other sites. Confirmed that Jilin City Mycoplasma pneumoniae clinical isolates the existence of 2063 loci of A- G gene mutation, indicating a possible is one of the reasons leading to the resistance of other strains, no mutation, indicating that there may be other mechanisms exist. Conclusions: 1. MP is an important pathogen of pneumonia in children with respiratory tract, no obvious gender differences and seasonal differences, but there are obvious age characteristics, good hair in infants and preschool children. 2. Rapid identification of 2 mycoplasma culture and drug sensitive test of simple operation, rapid, specific, easy to collect specimens, direct drug susceptibility results can guide the clinical rational use of drugs, the detection result can meet the clinical demand, can provide scientific basis for clinical diagnosis and treatment. 3. 16 SrRNA gene sequence because of its sensitivity, stability becomes the determining whether infection of mycoplasma detection technology. 4. The Jilin urban district 3 MP resistant to macrolides serious situation, one of the primary mechanism of resistance to 23 SrRNA V area of A2063 point mutation.
