Effect And Mechanism Of VEGFin The Process Of IL-1β Opening Blood-brain Tumor Barrier

Objectives To study the effect of IL-1β on protein expression of vascular endothelial growth factor, plasma membrane microcapsule structure protein caveolin-1 and caveolin-2 on glioma cells, preliminary discussion the possible mechanism of IL-1β opening blood tumor barrier.Methods Establish the tumor barrier model by transwell in vitro.Immune fluorescence method and Western blot method was used to detect the VEGF protein expression level after different doses(0.1 ng / ml, 0.25ng/ml and 0.5ng/ml, 0.75ng/ml and 1ng / ml) IL-1βtreated C6 glioma cells.The effect of IL-1β on the expression of caveolin-1 and caveolin-2 in C6 glioma cells was dynamically monitored by western blot.Transmembrane resistance method assessment the permeability of blood tumor barrier after IL-1β.Results The tumor barrier model was successfully established by transwell in vitro.When IL-1β was treated the model of blood tumor barrier, the expression of VEGF increased, and reached the peak at 60 min, and recovered to the initial state at 120 min.The transmembrane resistance of the blood tumor barrier model was the lowest at 60 min,at the same time, the transmittance of sodium fluorescein was the highest. In addition, our results also found that, the protein expression of cytoplasmic vesicles microbubble structural protein caveolin-1 and caveolin-2 in brain microvascular endothelial cells reached peak at 60 min, subsequently reduced and returned to non drug state at 120 min.Conclusions IL-1β increased blood tumor barrier permeability, the effect may be IL-1βUp-regulated the protein expression of caveolin-1 caveolin-2 by VEGF, thereby causing the opening of the blood-tumor barrier.
