Objectives By observing the expression of h MLH1,h MSH2 and h MSH6 protein in colorectal cancer and normal mucosal tissue. Analyze the relationship between the expression of three proteins in colorectal cancer clinical stage clinical parameters,such as the degree of differentiation,etc.And to investigate the expression of three proteins in colorectal cancer prognosis and the feasibility of tumor markers.Methods Collected from September 2012 to September 2014 during the Affiliated Hospital of North China University of Science Technology,General Surgery,72 cases with complete clinical and pathological data of patients with CRC who underwent radical resection specimens obtained paraffin-embedded cancer tissue cut from the Department of Pathology.Immunohistochemical SP method was used in 72 patients with CRC tissues and 26 cases of normal colorectal tissues h MLH1,h MSH2,h MSH6 protein expression were detected;c2test,Spearman correlation analysis to analyze h MLH1,h MSH2 and h MSH6 expression and protein deficiencies clinicopathological features of the patient’s sex,age,tumor location and tumor grade,lymph node metastasis.Results 1 In 72 cases of CRC tissues, there are different degrees of h MLH1 expression were missing for 53 cases,while there are 19 cases of normal expression,h MLH1 protein expression was 73.6% loss(53/72);and 26 cases of normal colorectal tissues,h MLH1 protein expression was lost 11.5%(3/26),h MLH1 proteins have relatively significant difference(c2=8.957,P<0.001) in the loss rate of CRC tissues and normal colorectal tissue.CRC in the absence of h MLH1 protein expression in tumor diameter ratio,degree of differentiation,Dukes stage,lymph node metastasis and distant metastasis(P<0.05),but has nothing to do with gender and age(P>0.05).2 In the 72 cases of CRC tissues tissues showed different degrees of h MSH2 protein expression in 47 cases of missing persons,and h MSH2 protein expression was normal in 25 cases,loss of expression of h MSH2 protein in CRC was 65.3%(47/72).The absence of h MSH2 protein expression in normal colorectal tissue was7.7%(2/26),h MSH2 protein in CRC tissues and normal colorectal tissue loss rate was statistically significant(c2=9.352,P < 0.001). h MSH2 protein expression loss rate of tumor diameter in CRC tissue differentiation,Dukes stage,lymph node metastasis and distant metastasis(P<0.05),and gender and age(P>0.05).3 In the 72 cases of CRC tissues,different levels of h MSH6 protein expression were missing for 31 cases,while normal h MSH6 protein for 41 cases,h MSH6 protein expression loss was 43.1%(31/72).h MSH6 protein in 26 cases of normal colorectal tissues missing was 3.8%(1/26),CRC tissue and normal colorectal tissue h MSH6 protein deletion rate was statistically significant(c2= 4.137,P<0.05).lack of expression of the degree of tumor differentiation h MSH6 protein,Dukes stage and lymph node metastasis(P < 0.05),but not with gender,age,tumor size and distant metastasis(P > 0.05).4h MLH1 、 h MSH2 protein expression in CRC tissues deletion rates were positively correlated(r= 0.295,P<0.05).h MLH1,h MSH6 and h MSH2, h MSH6 protein expression deletion was no significant relationship(P>0.05).Conclusions 1 In CRC tissues,The loss rate of h MLH1,h MSH2 and h MSH6 expression are higher than normal colorectal tissue.2CRC incidence,development and h MLH1,h MSH2, missing h MSH6 of expression.3 In colorectal cancer h MLH1, h MSH2 protein expression deletion tumor size, degree of differentiation, Dukes stage,lymph node metastasis and distant metastasis; h MLH1, h MSH2 protein expression was positively correlated deletion in colorectal cancer tissues; in colorectal cancer tissue expression of the extent of missing h MSH6 protein with tumor differentiation, Dukes stage, lymph node metastasis. h MLH1, h MSH2 and h MSH6 of colorectal cancer may be used as a new tumor marker.
