Objective This experiment aims to study the investigation focus on expression of c-jun in arsenic trioxide inhibited proliferation, induced apoptosis of adenoid cystic carcinoma-2(ACC-2) cells in vitro.The goal is to explore feasible clues on treatment of adenoid cystic carcinoma.Methods(1)The experimental groups were given various concentration of As2O3, while control groups without. Morphological changes were observed by inverted microscope.(2) RT-PCR method was used to investigate c-jun genetic changes in m RNA level in experimental and control groups;(3)Immunohistochemical method was applied to investigate Jun protein expression after As2O3 treatment. Using SPSS19.0 statistical analysis package to carry on the data processing, inspection level for a = 0.05.Results(1) Microscope observation: ACC-2 cells growth is slow, adherent ability decline in experimental group; As2O3 can inhibit the proliferation of ACC-2 cell and induce cell apoptosis, which was time-dependent and concentration-dependent.(2) RT-PCR:The results show that c- jun m RNA express the highest amount in the control group.With the increase of concentration of As2O3 and culturing time, the expression of c-jun m RNA decreased obviously, reach its bottom in 6μmol/L concertration at 48 h. The difference was statistically significant(P = 0.001).(3) Immunohistochemistry:The results show Jun protein expression in the nucleus with brown-yellow or brown,not in cytoplasm. Jun protein is highly expressed in ACC-2 cells,and its expression decreased in ACC-2 cell treated with As2O3 in a time-and dose-dependent manner.The difference was statistically significant(P = 0.008).Conclusion(1)As2O3 can inhibit the growth of ACC- 2 cells in vitro and induce cell apoptosis;(2) The expression of c-jun m RNA and protein has obvious specificity in ACC-2 cells in vitro;And As2O3 can change the level of expression of c-jun m RNA and protein level, which was time-dependent and concentration-dependent;(3) c-jun gene play an important role in the apoptosis of ACC-2 cell induced by As2O3,which may provide a good base on theory for clinical application of As2O3 on ACC.
