ObjectiveWhen jaw keratocystic odontogenic tumor(KCOT) was found, itwas always very large. Marsupialization was a minimally invasive method to treat large KCOT, but it need a long period of treatment.Wnt signaling pathway was closely related to human tumor.The expressions of Wnt signaling pathway genes in KCOT were investigated before and after marsupialization in the study. Then Wnt signaling pathway inhibitor was used to treat cases with KCOT to shorten the period of marsupialization and the expression of Wnt signaling pathway genes were detected to explore the role of Wnt signaling pathway genes in marsupializationof KCOT.Methods and MaterialsFresh tissues of KCOT were obtained and the expressions of Wnt signaling pathway genes were detected by gene chip. Fifteen cases with KCOT were treated with marsupialization, six cases washed with EGCG(Wnt signaling pathway inhibitor) and nine caese washed with normal saline(NS). The cavity of KCOT was gradually decreased after marsupialization and the period of treatment was observed. When the size of the cavity less than 2cm×1cm, the secondary operation was done to take off the left cyst wall. The fresh specimen was obtained and the expressions of Wnt signaling pathway genes were detected by gene chip.Results1. The expressions of Wnt signaling pathway genes, FZD3 and MAPK10, were upregulated in jaw KCOT.2.Jaw KCOT was shrunk gradually after marsupialization and secondary operation was taken in 9 months with EGCG and in 16.2 months with NS. EGCG had obviously shortened the clinical treatment period.3.The expressions of FZD3 and MAPK10 were no upregulation in the left cyst wall after treatment with EGCG, that similar to normal mucosa. While the expressions ofFZD3 and MAPK10 were still upregulated in left cyst wall after treatment with NS,which lower than the cyst before treatment, there was no significant difference between them.Conclusions1.The expressions of FZD3 and MAPK10, Wnt signaling pathway genes, were upregulated in jaw KCOT.2. EGCG, Wnt signaling pathway inhibitor, could obviously shorten the period of marsupialization of KCOT.3.The expressions of FZD3 and MAPK10 were sharply decreased in left cyst wall after marsupialization with EGCG, similar to normal mucosa.Wnt signaling pathway genes might play a role in the formation and treatment of jaw KCOT.
