Objective: To study the protective effect of Ashitaba Chalcone(AC) on acute radiation-injured mice.Method: Fifty healthy kunming mice with half males and females were randomly divided into 5groups after feeding in our laborotary for one week, including control group, radiation injury group, low dose of AC group, middle dose of AC group and high dose of AC group, per group consisted of five mice. The above each experiment group was given by gavage once a day for five days with 4mg/kg AC for low dose group, 20 mg/kg AC for middle dose group, 40 mg/kg for high dose group, while the control group and radiation injury group were given saline by gavage. At sixth day, all experiment group were irradiated with 4 Gy x-ray once except blank control group,then all mice were given different dose of AC as usual for the next five days. At the sixth day, all mice were sacrificed to detected the following indicators:1. Detecting the body weight of mice at the day before gavage, the day before irradiation and the sixth day after irradiation with precision electronic balance.2. Determination of T-AOC, MDA and SOD of liver by spectrophotometric method.3. Determination of Bcl-2 and Bax protein expression of liver by Immunohistochemical method.4. Detecting proliferation activity of spleen cell by MTT method.5. Determination of the bone marrow polychromatic erythrocyte micronuclei ratio.Results:(1) The body weight of mice exposure to 4 mg/kg AC group was 24.396 ± 1.388 g and24.792 ± 0.919 g respectively at the day before irradiation and the sixth day after irradiation; 20mg/kg AC group was 24.375 ± 1.247 g and 25.578 ± 0.916 g; irradiation injured group was 22.883± 1.252 g and 22.778 ± 0.829 g. The difference of body weight in each experiment group had statistically significance( p<0.05).(2) The T-AOC and SOD activity of mice exposed to 40 mg/kg AC was 3.901± 0.304 U/ml and 71.938± 1.051 U/ml respectively, and significantly higher than irradiation injured group(p<0.05); the MDA activity was 1.864± 0.324 nmol/ml, significantly lower than irradiation injured group(p<0.05).(3) The Bcl-2 and Bax protein levels of liver from single irradiation injured group was 56.383 ± 2.282 and 72.440 ± 0.455 repectively, while 40mg/kg AC group was 76.975 ± 1.847 and 48.782 ± 0.929, the differences of Bcl-2 and Bax protein in both single irradiation injured group and 40 mg/kg AC group were statistically significantly(p<0.05).(4) The proliferation activity of spleen lymphocytes was 0.534 ± 0.179 and 0.607 ±0.031 respectively from irradiation injured group and 40 mg/kg AC group, and the difference had stastically significance(p<0.05).(5) The micronuclei rate of polychromatic erythrocytes in mouse bone marrow was 8.667 ± 0.543 ‰ and 18.912 ± 0.525 ‰,and the difference had stastically significance in above two experiment group(p<0.05).Conclusion: Ashitaba Chalcone could suppress the decreace of T-AOC and SOD acticity and increase of lipid peroxidation level, raise the Bcl-2 and Bax protein expression, improve the proliferation activity of spleen lymphocytes, and reduce the bone marrow polychromatic erythrocyte micronuclei ratio to protect mice from serious irradiation injury.
