| Objective: By detecting EPOR expression and observe apoptosis of brain tissue to explore the protection mechanism of erythropoietin for intrauterine inflammation neural rat.Method: Select 12 Wistar rats that were only 15 days of pregnancy, according to the proportion of 3:1 randomly divided into inflammation group(n = 9) and the control group(n = 3). Inflammation group was given LPS by celiac injection, and control group was given amount of saline solution. HE dyeing pathologic examination of brain and placental tissue was given after two groups of rats delivery. Inflammation group remaining live young mice were randomly divided into intrauterine rh EPO treatment group(n = 20) and intrauterine inflammation group(n = 20), and normal saline control group remaining live young mice in the control group(n = 20).Among them, intrauterine rh EPO treatment group rats was given rh EPO(5000 IU/Kg) by celiac injection after birth immediately, the rest of the two groups were given amount of saline solution. 8 groups of young rats were given brain perfusion after birth, 3 days, 7 days, and 14 days with ELISA method to detect brain EPOR protein expression.Result: 1.Placental tissue of inflammation group was hyperemia and edema,accompanied by a large number of neutrophils.2. Brain structure of inflammation group was loose, disordered arrangement of nerve fibers. Brain structure of control group was clear, nerve fiber line and tidy. Brain structure of intrauterine rh EPO treatment group was between the other two groups.3. Intrauterine rh EPO treatment group compared with intrauterine inflammation group,physiological saline control group, the EPOR protein increased significantly by the age of3, 7 days of young rats, and the differences between the groups were statistically significant(P < 0.05).Conclusion: 1.By celiac injection of LPS can build intrauterine inflammation animal model.2. Newborn rats after celiac injection of rh EPO can inhibit nerve cell apoptosis, and has therapeutic effect on brain injury.3. Giving early infected rats celiac injection of rh EPO can increase the expression of brain EPOR, by increasing the expression of brain EPOR to suppress neural cell apoptosis may be one of its play to the role of neural protection mechanism. |
