Objective:Through the discussion of GRP78 occur in the development of diabetic rat detrusor cell endoplasmic reticulum stress lesions, especially mechanism of detrusor smooth muscle cell apoptosis in the process, reveal the GRP78 in diabetic rat detrusor cell endoplasmic reticulum stress lesions occurred development in the dynamic evolution rule changes, for the study of the etiology of DCP provide important experimental basis.Methods:In this study, we selected GRP78 as the marker of Sprague-Dawley, and healthy cleaning grade adult female ERS rats were randomly divided into 4 groups and 1 groups of normal control group. The diabetic group was injected with intraperitoneal injection of 16.7mmol/L(STZ) 50mg/kg, the blood glucose concentration of tail vein after 72 hours, and the model was successful.. Respectively at 4 weeks, 8 weeks, 12 weeks and 16 weeks were rats, resection of bladder tissue, and removal of surrounding fat and connective tissue, respectively by TUN EL method for the determination of detrusor cell apoptosis rate and electron microscope to observe the morphological changes of endoplasmic reticulum of detrusor cells. Relationship was detected by RT-PCR and Western blot dynamic study of diabetic rat detrusor apoptosis level of GRP78 expression, and all the data of the statistical analysis. Result:Diabetic rats appeared polydipsia, polyphagia, increased urination, significantly thin, yellow hair hardens, glossiness decreased, drowsiness unresponsive to external conditions, individual limbs ulceration in diabetic rats, not sensitive to pain, the rats in the control group of normal diet; the apoptosis rate increased in diabetic groups with the increase of time, the trend of significantly increased at 12 W, the apoptosis trend was time dependent; electron microscopy in the endoplasmic reticulum around the nucleus of detrusor smooth muscle cells in diabetic group a large number of endoplasmic reticulum increased volume, disordered, white irregular vacuoles, membrane attached ribosome; RT-PCR showed that, 4W group and 8W group was significantly higher than that of the control group, 12 W group, the expression level of the highest, is 8.87 times higher than that of control group, 16 W group and 12 W expression level is decreased. Western- blot test showed that the expression level of 4W protein in the GRP78 group was higher than that in the control group, the expression level of 8W group was the highest, and it was 1.45 times of the control group, while the 12 W group and 8W group were significantly decreased compared with the 16 W group. Conclusion:DCP detrusor smooth muscle cell apoptosis and endoplasmic reticulum stress is closely related to including GRP78 expression level changes to detrusor smooth muscle cell apoptosis plays an important role in the pathogenesis and with the progression of diabetic, GRP78 and DCP have obvious correlation. By studying the relationship between DCP and GRP78, we can provide a new potential target for the treatment of diabetic bladder and its potential therapeutic target.
