| Objective: To probe into the effects of the apoptosis of granulose cells regulated by SIRT1 on ICSI outcomes.Methods: Gene expression of SIRT1 and FOXO3 a were performed by real-time PCR, immunocytochemical staining and western-blot. Apoptosis of granulosa cells was detected by flow cytometry with the expression of Caspase-3. The follicular fluid analysis was performed by chemiluminescence assay.Results: 1. Significant correlation was found between SIRT1 expression and apoptosis on human granulose cells in cumulus during ICSI treatment(P<0.05).2. The apoptosis rates of granulose cells were elevated with the increase of SIRT1 expression. The number of aspirated follicles, oocytes obtained, oocytes available for ICSI, oocytes fertilized, cleavage fertilized oocytes, high quality embryos, blastocyts and frozen blastocysts for ICSI patients were decreased significantly(P<0.01).3. The significant correlation of m RNA expression between SIRT1 and FOXO3 a was found in cumulus during ICSI treatment(P<0.01).4. The significant correlation between SIRT1 expression in granulose cell and E2 level in follicular fluid was found during ICSI treatment(P<0.01)Conclusion:ICSI outcomes were affected by apoptosis of human granulose cells which was regulated by SIRT1 during the ICSI treatment, and the apoptosis of granulose cells was induced by SIRT1 via FOXO3 a regulations. |
