Objective:Most animal models of uveitis experimental research are EAU or EIU. Compared to two kinds of model : long time EAU model, but chan ge of anterior segment is not obvious. The EIU model inflammatory respon se is fast withshort duration,and anterior segment reaction obviously.The EIU model is beneficial to clinical observation and inflammation grading.In view of the characteristics of two kinds of animal models,we tried to establish an model which is observed convenient and longstanding inflammatory.methods:, we use humanretinal soluble antigen(s- Ag) peptide fragments(P35) and salmonella typhi endotoxin to induce rat model in lewis.The clinic al features, histology, and serum inflammatory factors change has carried on the detailed observation and measurement.Then investigate the ocular inflam mation and pathological manifestation,and analysis the TNF-α and IFN-? changes in serum concentrations.Results:(1) The anterior segment reaction in HS- Ag P35 group was light,but the posterior segment observed widely retinal detachment.Even if in the peak period,we could not seen anterior chamber empyema, pupil adhesion and obvious lens cloudy.The eye clinical signs was out of step with its tissue pathological changes. However the HS- Ag P35+ LPS group had consistent of clinical and tissue pathological changes.(2) TNF-α level of HS-Ag P35+LPS group was bimodal, reached the peak on the 2th and 4th day after the second immunization. HS-Ag P35 group reached a peak on the 6th day.There are no difference between HS-Ag P35 + LPS and HS-Ag P35 group in 6th TNF-α level.(3) Both two groups of experimental rats could detect IFN-?,and higher than normal rats. In the 10 th days after second immunization they had peak with little differences.Conclusion:We choose HS- Ag P35 to establish rat models, In order to improve the anterior segment inflammation reaction in EAU,we use endotoxin vitreous injection at the same time.And set up a new kind of rat models with strong anterior segment inflammation reaction and long duration.The experiment methods of this kind of models are simple,and easy to observation and evaluation. This model is especially suitable for new drugs’ clinical effectiveness detection and evaluation.
