BackgroundSpleen is the largest immune organ in the body. Spleen injury is one of the most common form of abdominal trauma. Splenectomy may not affect the safety of life in a short time, but the immune level of patients will be reduced significantly, and this will increase the susceptibility of serious infections such as overwhelming post-splenectomy infection(OPSI), septicemia and meningitis. Especially in infant, splenectomy will increase the opportunity to infectious complications. In addition, the cell immunity and humoral immune will become disorder and affect the occurrence and development of tumor after splenectomy. Then, for tumor injury patients, we should rescue their life and try our best to reserve the immune function of spleen as far as possible. Hence, splenic auto-transplantation is of great significance. ObjectiveIn this study, we compare the concentration of blood immune index, regenerate status of blood vessels and nerves by immunohistochemistry and ELISA in different rat models at different time. Try to clarify the rules of splenic autotransplantaion and try to provide a theoretical basis for spleen tissue transplantation.Methods1. Rat models of total splenectomy,and splenic auto-transplantation were established.2. The blood vessels and nerves at different time points after operation were observed by immunohistochemistry.3. Ultrastructure of spleen cells at different time points after operation was observed by transmission electron microscope.4. Concentration of CTL、IL-2、LAK、NFКB、Tuftsin in the blood were measured by ELISA at different time points after operation. Results1. Spleen cells began to regenerate on the 14 th day after splenic auto-transplantation, and the cells’ structure were integrated and organelles were visible clearly on the 60 th day.2. Blood vessels began to regenerate on the 14 th day after splenic auto-transplantation, and widely distributed in spleen tissues on the 60 th day.3. Nerves began to regenerate on the 7th day after splenic auto-transplantation, and widely distributed in splenic tissue on the 60 th day.4. Transplanted spleen tissues undergo the progress of necrosis, regeneration and reconstruction. Normal spleen tissues’ structure were formed primarily on the 60 th day after operation.5. The concentrations of CTL, IL-2, LAK, Tuftsin in animal model blood increased gradually and get close to the normal level on the 60 th day after operation. ConclusionIn the splenic autotransplantation group, the transplanted spleen would go through the progress of necrosis and regeneration, the tissue structure returned to normal gradually, blood vessels and nerves regeneration step by step, red pulp, white pulp and fringe area and other characteristic splenic structure which approximate same as normal splenic corpuscle structure were formed. All above provides morphological basis of the spleen function. At the same time, related immune index level of blood were recovered, the result further proved that auto-transplantation of spleen into greater omentum after splenectomy is a simple and effective way to transplantation autologous spleen tissue in clinic. The method plays an important role in maintain the body’s normal immune function.
