Abstract/Summary:
Objective:To investigate the expression level of programmed cell death 5(PDCD5) in the serum of advanced non-small cell lung cancer(NSCLC), and to explore the correlation study of its sensitivity to platinum chemotherapy drugs.Methods: Collected 40 advanced non-small cell lung cancer patients confirmed by pathological examination and designated them as lung cancer group. Collected another 20 persons whose physical examination were healthy as normal group. Patients in lung cancer group were given platinum drugs combined with other chemotherapy drugs. The chemotherapy cycle was 21 days and curative effect evaluation was made after 2 cycles of treatment. According to efficacy they were divided into effective group and ineffective group.ELISA was used to detect the serum PDCD5 concentration in lung cancer group and normal control group, and its relationship with curative effect and clinical significance was analysed.Results:1. The serum PDCD5 protein level in the normal group was(1.163±0.293)ng/ml and lung cancer group was(0.831±0.352)ng/ml. The level of serum PDCD5 protein in the normal group was higher than the lung cancer group and the difference was statistically significant(t=2.292,P=0.028).2. 40 cases of patients with advanced NSCLC CR0 cases, PR23 cases, SD7 cases,PD10 cases, so effective group was 23 cases and objective response rate was 57.5%,ineffective group was 17 cases. Before chemotherapy the serum PDCD5 protein concentration in effective chemotherapy group was(0.850±0.216) ng/ml higher than the ineffective group(0.668±0.287) ng/ml, and the difference was statistically significant(t=2.292,P=0.028). After chemotherapy the serum PDCD5 protein concentration in effective chemotherapy group was(1.083±0.493)ng/ml higher than the ineffective group(0.572±0.142)ng/ml. After chemotherapy the PDCD5 protein level difference was dramatically increased between the effective group and the ineffective group, and the difference was significant statistically significance(t=4.980,P=0.000). After chemotherapy effective group PDCD5 protein level was higher than before, the difference was statistically significant(t=-2.540,P=0.021). After chemotherapy ineffective group PDCD5 protein level was lower than before, the difference was statistically significant(t=2.440,P=0.027).3. PDCD5 protein level was associated with lymph node metastasis and differentiated degree(P<0.05). It was no correlated with smoking history, gender, pathological type(P>0.05).Conclusion: 1. PDCD5 may be a new kind of inhibition of tumor growth and promote apoptosis gene and which expression levels reduce may relate to the occurrence of lung cancer.2.PDCD5 protein level in serum of patients with advanced NSCLC is expected to be a predictor of chemosensitivity. The expression levels of PDCD5 may affect the sensitivity to cisplatin chemotherapy in patients with lung cancer, which may prompt chemotherapy predictor to patients with advanced NSCLC. To provide a new direction formode of anticancer gene combined with chemotherapy.3. In the serum of patients with advanced NSCLC, PDCD5 protein levels correlated with lymph node metastasis and tumor differentiation. The down-regulation of PDCD5 expression level may be an adverse prognostic factor of advanced NSCLC patients.
