Objective: To establish an animal model of iron deficiency and observe general condition of these animals.Detecting the hematologic parameters: hemoglobin(Hb) concentration、red blood cell count(RBC)、mean corpuscular volume(MCV) 、 hematocrit(PCV) 、 mean cell hemoglobin(MCH) 、 red blood cell volume distribution width(RDW),ferritin(Fn),erythropoietin(EPO) and Insulin-like growth factor-1(IGF-1) during perinatal iron deficiency, and detecting the hormones level of endocrine gland during perinatal iron deficiency at the same time, and the phenomenon of low birth weight in newborn rats. To explore the newborn rats’ endocrine glands and liver whether or not have erythropoietin receptor expression, and to detect whether or not have been effected by iron deficiency of mother on erythropoietin and insulin-like growth factor-1 in liver. Initially attempts to complete the cultivation of hypothalamus, pituitary, adrenal gland in vitro,for the study of secretory function of hypothalamic-pituitary-adrenal axis(HPA) paving the way.Methods: 23 healthy female Wistar rats were selected randomly, one week after feeding adaptation, and randomly divided into the experimental group and the control group, 16 rats in each group. The rats in experimental group were fed with low iron diet(iron content:10mg/Kg) and the rats in control group were fed with normal diet(iron content:216mg/Kg), then begin the intervention. Four weeks after intervention,the rats were mated with healthy adult male rats and continued to feed each diet until delivery. The levels of hematological parameters, Fn, EPO, IGF-1 and other hormones which were secreted by the endocrine glands were detected during perinatal rats. After the offspring were born, the newborn birth and weight were observed. The endocrine glands of hypothalamus, pituitary and adrenal glands and livers of newborn rats were collected for tissue slice to detect the level of erythropoietin receptor expression; the livers of newborn rats were collected for tissue homogenate to measure the level of IGF-1 and EPO. In addition to culture the hypothalamus, pituitary and adrenal gland tissue in vitro, using recombinant human erythropoietin stimulate single endocrine glands cells. And detecting the the secretory of endocrine glands such as growth hormone(GH), growth hormone-releasing hormone(GHRH), Corticosterone(CORT),Epinephrine(EPI) and Adrenocorticotropic hormone(ACTH).Results: ①.Rat model of perinatal iron deficiency were established successfully. The level of Hb,MCV,MCH,PCV,Fn,GH and CORT in experimental group were statistically significantly lower than those in control group(P<0.05) during perinatal period;however the RDW and the level of ACTH、GHRH and EPI in plasma and liver homogenates in experimental group were statistically significantly higher than those in control group(P<0.05).②.The phenomenon of stillbirth was found in experimental group and the stillbirth rate of newborn rats in experimental group was significantly higher compared with the control group(P<0.05). ③.Compared with the control group, the level of nsulin-like growth factor-1 in tissue homogenate were lower in experimental offspring groups(P<0.05); Pituitary, adrenal glands and livers of newborn rats in experimental group have a little more erythropoietin receptor expression than control group. ④.The techniques of vitro culture in hypothalamus, pituitary, adrenal have been mastered initially ⑤.EPO stimulate the endocrine glands, the hormone secretion level of endocrine glands were no significantly between the experimental group and the control group.Conclusions: 1. Perinatal iron deficiency in rat model is established successfully.2. Perinatal iron deficiency not only affects parental Fn,EPO, GH, IGF-1, GHRH, ACTH,EPI and CORT, also has a certain influence of birth weight on offspring.3. The techniques of vitro culture in hypothalamus, pituitary, adrenal have been mastered initially. It is the experimental base of the study of secretory function of hypothalamic-pituitary-adrenal axis(HPA) in the further.
