Objective :Studying on the different levels of gene expression in Pup-proteasome system(PPS) of different drug-resistance clinical isolates of mycobacterium tuberculosis(hereinafter referred to as the MTB), and exploring the correlation between Mycobacterium tuberculosis Pup-proteasome system(PPS)and drug-resistance of production and mechanism with spread in Xinjiang region.Methods:Culturing INH-MTB, RFP-MTB, SM-MTB, EB-MTB, MDR strains at Original drug resistance Medium, low concentrations of drug Medium and high concentrations of drug Medium to the logarithmic phase, and extracting total RNA from each group of MTB. Using SYBR Green I real-time PCR method to detect the Pup gene, Dop gene, Mpa gene and Paf A gene expression level in different concentrations of drug-resistance MTB in each group of MTB.Results:(1) In original state, Compared with the drug-senstive clinical isolates of MTB,m RNA expression level of Pup gene was down-regulated in INH-MTB, EB-MTB, MDR; m RNA expression level of Dop gene was higher in INH-MTB, SM-MTB, EB-MTB; m RNA expression level of Paf A gene was higher in INH-MTB, RFP-MTB, SM-MTB, EB-MTB and MDR; m RNA expression level of Mpa gene was down-regulated in INH-MTB and MDR, however,m RNA expression level of Mpa gene was higher in SM-MTB, the difference was statistically significant(P<0.05);(2) Under the influence of anti-TB drugs(INH, REP, SM, EB), compared with the original state of drug-resistant MTB strains,at low concentrations of Anti-MTB drugs condition,the expression of Pup gene in INH-MTB, RFP-MTB, SM-MTB and MDR strains group was down regulated; the expression of Dop gene, Paf A gene and Mpa gene was significantly up regulated;the difference was statistically significant(P<0.05);(3) Under the influence of anti-TB drugs(INH, REP, SM, EB), compared with the original state of drug-resistant MTB strains, at high concentrations of Anti-MTB drugs condition, the expression of Pup gene in INH-MTB, RFP-MTB,SM-MTB, MDR strains group was down regulated; the expression of Dop gene, Paf A gene and Mpa gene was up significantly regulated; the difference was statistically significant(P<0.05);(4) Compared with Low concentrations of anti-TB drugs clinical isolates of MTB, the expression of Pup gene, Dop gene, Paf A gene and Mpa gene in high concentrations of anti-TB drugs clinical isolates of MTB on(INH, RFP, SM)pressure was much higher; the difference was statistically significant(P<0.05);Conclusions:(1)In original state, there are different in m RNA expression in Pup-proteasome system(PPS)of the drug sensitive isolates of MTB and INH-MTB, RFP-MTB, SM-MTB, EB-MTB, MDR strains;(2) Under the influence of anti-TB drugs(INH, REP,SM, EB), the m RNA expression levels of genes in Dop,Paf A, Mpa was significantly higher than the original state; however,m RNA expression level of gene in Pup gene was lower than the original state.
