Objective: The aim of this study is to evaluate the mean of percutaneous coronary occlusion to establish Chinese mini-pigs acute myocardial infarction(AMI) model,and observe dynamic changes of serum cardiac fatty acid binding protein(HFABP) in order to provides reliable clinical data for the early diagnosis of AMI,and to explore pathological changes of myocardial infarction for the best timing of revascularization.Methods: 16 mini- swines were enrolled stury because of similar to human coronary artery. The obtuse marginal branches(OM branch) of 16 Chinese mini-swine wered blocked by gelatin sponge via microcatheter under general anesthesia through seldinger right femoral artery puncture after coronary angiography. AMI animal models were set up successfully after distal blood flow disappear and ST-segment of ECG display upward.At the same time,pull out the femoral artery sheath into the subclavian vein cannula for postoperative bleeding and blood.Serum H-FABP concentrations of before AMI、0.5Hour after AMI,1Hour, 2Hour, 3Hour, 4Hour, 6Hour, 8Hour, 10 Hour, 12 Hour, 24 Hour, 48 Hour point were observed with the mean of Ellisa after successful of AMI animal models to observe serum H-FABP dynamic change.Later, infarct myocardium was fixed 4% formaldehyde after lavaged by NS in HE-staining,Pathological changes and repair process of infarct myocardium after AMI 1.0d, 3.0d, 5.0d, 7.0d, 10.0d was observed on different days.Results: The OM branch of 16 Chinese mini-pigs were successful blocked by gelatin sponge.Serum H-FABP in the 0.5h began to increase after AMI, 8h reached its peak,and gradually fell back to normal after 24h(F=4.460,P<0.001). At 1d after AMI,the myocardial necrosis outline remain with strongly eosinophilic cytoplasm, nucleus deformation and mainly granular cytoplasm.At 3d after AMI,the myocardial necrosis outline vaguely remain,with a large number of inflammatory cell on the edge of myocardial cell.At 5d after AMI, nucleus of myocardial cell fragmented,with a little inflammatory cell.At 7d after AMI, a large number of granulation tissue development on the edge of myocardial cell,with a small amount offibrous tissue.At 10 d after AMI, a large number of scar tissues had displaced granulation tissue.Conclusion: 1. It is scientific, practical, successful to copy mini-swine AMI model by percutaneous coronary occlusion. 2. Serum H-FABP in the 0.5h began to increase after AMI, 8h reached its peak,and gradually fell back to normal after 24 h.Monitoring of the dynamic changes of serum concentrations of H-FABP can provide clinical data for early diagnosis and treatment of patients with AMI. 3. At 7-10 d after AMI,Revascularization is relatively safe because of complete repair of necrotic myocardial cell.
