AbstractObjective: Extract oxalis active ingredient with different ways, analsye them and observe their different components’ s protective effect on the liver damage in rats caused by the a-naphthyl isothiocyanate.Methods: 1. Extract oxalis active ingredient with different ways, they are espectively: ①immerse the Oxalis powder in the proper amount of distilled water and place them on a magnetic stirrer. After heated, stirred, then filtered, concentrated and freeze-dried to obtain oxalisⅠ. ②Add with hydrochloric acid, ethanol extraction, soak or heat them with water and stir intermittently. After the end of the reaction, it was filtered and concentrated to a paste to get oxalis Ⅱ. ③Add ethanol, heat at reflux for extraction, wash with diethyl ether, separate upper solution, add dropwise a solution of sodium bicarbonate, and extract twice with trichloride, take aqueous layer, hydrochloric acid solution is added and oxalis Ⅲ is gotton after the the precipitate and the dry. Analyse the structure of the extracted ingredients to know its main compents and defect its antioxidant, superoxide radical scavenging effect.2. The oxalis Ⅰ, oxalis Ⅱand oxalis Ⅲ were given orally to rats, observe oxalis’ active ingredients effects on the liver injury in rats, their rectal temperature, biochemical indicators,NO, etcResults: Extract oxalis active ingredient with three different ways, they are espectively:oxalis Ⅰ, oxalis Ⅱand oxalis Ⅲ. After structure analysing, their main active ingredients are polysaccharide compound, anthocyanin compounds and flavonoids with different characters colors and different effects on the antioxidant and liver injury intervention experiments. Of which the oxalis Ⅲ’s antioxidant is most powerful.Injury model of rat liver oxalis each active ingredient in the a- naphthyl isothiocyanate(ANIT) induced a certain intervention, experimental rats appear poisoned ANIT weight plummeted, metabolic disorder, there loss fall, listlessness and other symptoms, indicating successful model. Each active ingredient has a certain intervention to the liver injury of the rats caused by the a-naphthyl isothiocyanate. In the experiments, the ANIT poisoned rats loseweight sharply, appear metabolic disorder, lose appetite and are listless, all indicationg the seccess of creating the model. But the adding the three solutions of the extracted oxalis will relive the symptoms to a certian degree. Compared with model rats, the oxalis can obviously increase the weight and decrease the rectal temperature. The oxalis Ⅱhas no effects on the rats’ weight. In the experiments of measuring Alkaline phosphatase(ALP), alanine aminotransferase(ALT), total choline(TB), aspartate aminotransferase(AST), we can clearly see that each model group’s ALP, AST, ALT, TB in serum was significantly higher. But after the intervetion of the oxalis, the ALP, AST, ALT, TB in serum are lower than the model significantly. Of the indexes content, oxalisⅢ was significantly lower than oxalis Ⅰ and Ⅱ.This shows that the oxalisⅢ protects liver damage maximum and can significantly improve liver damage.3. The experiment shows that in the blood of model liver, nitric oxide was found and in red blood cells, nitric oxide(NO) were significantly increased. NO elevated levels can lead to increased oxidative stress and thus increase the peroxide, affect liver function. Experimental results show that the all the oxalis ingredinets can make model rats’ NO decreased.Conclusions:1.The extraced ingredients from the oxalis are respectively oxalis Ⅰ, oxalis Ⅱand oxalisⅢ. Their mian active compoments are polysaccharide compound, anthocyanin compounds and flavonoids.2.This study shows that oxalis is antioxidant and has a protective effect on the liver damage in rats caused by a- naphthyl isothiocyanate. These effects are closely related to the active ingredient and structure.
