Objectives:To investigate the effect of emetine on proliferation and apoptosis in human renal carcinoma 786-0 cells and the mechanism involved, and to provide a theoretical basis for emetine applied in the clinical research of renal cell carcinoma.Methods:1.Renal cell carcinoma 786-O cells were cultured in vitro.We selected the logarithmic growth phase cells.Then,experimental groups were treated with different concentrations of emetine at 24,48,72h; normal cell control group was only added medium; solvent control group was added 0.16‰DMSO,which were detected by MTT assay in order to test the effect of drugs on cell proliferation.2. We analyzed the apoptosis rates of 786-O cells by Flow C ytometry.3. PI3 K and Akt m RNA relative expression levels in 786-O cells were detected by RT-PCR.4 Relative expression levels of total Akt protein and phosphorylated Akt( pAkt) protein in 786-O cells were detected by Western-blotting.Results:1 Emetine could significantly inhibit 786-0 cells proliferation compared with the control group. The inhibition increased, with the increase of more than 25 n M concentrations and the extension of time.The effect had a time-and dose-dependent manner(P <0.01).2 The rates of apoptosis were significantly increased compared with the control group, the differences had a statistical significance(P <0.01), the rates ascended as doses increased.3 PI3 K m RNA expression levels were significantly decreased compared with the control group(P <0.01), the levels decreased as doses increased. Meanwhile, Akt m RNA expression levels showed no significant change,the results had no statistical significance(P >0.05).4 p-Akt expression levels were significantly decreased compared with the control group(P <0.01), and negatively correlated to the dose.While total Akt protein expression levels showed no significant change, the results had no statistical significance(P >0.05).Conclusions:1 Emetine can inhib it renal cell carcinoma 786-O cells proliferation in vitro,the effect has a time-and dose-dependent manner.2 Emetine can induce renal cell carcinoma 786-O cells apoptosis in vitro.3 Emetine can down-regulate the expression levels of PI3 K m RNA and p-Akt protein,but it has no effect on the expression levels of Akt m RNA and total Akt protein.4 Inhibiting PI3K- Akt signaling pathway may be the important mechanism of emetine anti-renal cell carcinoma.
