Objective:Research KLF4 gene and MMP9 gene in hepatocellular carcinoma and adjacent liver tissues, to investigate K LF4 gene regulation MMP 9 gene the impact of expression changes in of liver cancer cell invasion and metastasis,to clarify molecular mechanisms of HCC invasion and metastasis,clear the molecular regulatory network of liver invasion and metastasis, to providing a theoretical basis for the treatment of liver cancer invasion and metastasis. Methods:1. Collect the tissue samples of hepatocellular carcinoma and the corresponding, detect the expression of MMP9 gene and KLF4 gene in 50 cases of hepatocellular carcinoma tissue and the corresponding tissue by immunohistochemistry, q RT-PCR and WB.2. Recombinant plasmid,increased expression of KLF4 gene in Hep G2 cells,to detect the expression of m RNA and protein of MMP9 gene.3. Transfected Hep G2 cells,detect the changes of invasion and metastasis by Transwell invasion assay, scratch test.4. Statistical analysis was performed using SPSS19.0 statistical software, the measurement data of normal distribution, the Fang C haqi, in order to c ±s said, comparison between groups using t test, analysis of multiple groups was compared with single factor analysis of variance, P<0.05, the difference was statistically significant. Results:1.q RT-PCR test results detection K LF4 m RNA expression of hepatocellular carcinoma was significantly lower than that of the corresponding tissue adjacent to carcinoma(P <0.05); MMP9 m RNA expression hepatocellular carcinoma was significantly higher than tthat of the corresponding tissue adjacent to carcinoma(P <0.05). Western blot test results found KLF4 protein expression of hepatocellular carcinoma was significantly lower than that of the corresponding tissue adjacent to carcinoma(P <0.05);MMP9 protein expression of hepatocellular carcinoma was significantly lower than that of the corresponding tissue adjacent to carcinoma(P <0.05).2.Immunohistochemical test results detection KLF4 prote in in 76%(38/50) of the cases showed low expression in HCC,While only 10%(5/50) showed low expression in cancerous tissues.;MMP9 protein in 84%(42/50) of the cases showed high expression in HCC, While only 12%( 6/50) showed high expression in cancerous tissues.3.q RT-PCR and WB test results detection that expression of KLF4 m RN A and protein in the genome of KLF4 compared with the blank control group and the ineffective treatment group increased significantly(P <0.05), While in the control group and treatment group KLF4 between the invalid m RNA expression had no significant difference(P>0.05). In addition, MMP9 m RN A and protein expression decreased with the increase of KLF4 expression.4.Transwell invasion assay, scratch test results showed: over expression of KLF4 cells invasion ability compared to the blank control group and the treatment group significantly reduced invalid(P <0.05); scratch test results showed that: over expression of Hep G2 cells in KLF4 group than that in blank control group wound healing rate and cell scratch healing rate of treatment group is obviously decreased(P <0.05). Conclusion:1. In hepatocellular carcinoma, KLF4 gene expression of hepatocellular carcinoma was significantly lower than that of the corresponding tissue adjacent to carcinoma; MMP9 gene expression hepatocellular carcinoma was significantly higher than tthat of the corresponding tissue adjacent to carcinoma.2. Up regulation of K LF4 gene expression in liver cancer cells lead to the decrease of MMP9 gene expression, and then inhib it the invasion and metastasis of liver cancer cel s.
