Objective: To investigate the expression of mi R-21 and PTEN m RNA in bone marrow mononuclear cells(BM-MNCs) of acute myeloid leukemia patients, and analyze the correlation between mi R-21 and PTEN m RNA and each of their correlation with the clinical features of the patients, and evaluate the clinical significance of the expression levels of mi R-21 and PTEN in AML patients.Methods: 1、Collected total 80 cases of BM samples with AML patients, including 43 cases of new diagnosed patients(according to FAB classification as an example, M1 of 5 examples, M2 of 16 cases, M3 of 7 examples, M4 of 5 examples, M5 of 7 examples, and M6 of 3 cases);13 cases of refractory and relapse patients, 24 cases of complete remission(CR) patients. 20 cases of non-neoplastic patients were used as controls.2、BM-MNCs were isolated from BM samples and extracted total RNA, then were reversed transcription into c DNA. Using the method of real-time fluorescent quantitative PCR to detected the relative expression of mi R-21 and PTEN m RNA. The formula 2-△ Ct was used to calculate relative gene expression values..3、Using SPSS17.0 analyze the data.Compare each groups’ differents with Mann-Whitney U test, analyze its correlation with the clinical features of the patients and the correlation between mi R-21 and PTEN m RNA with Spearman test.(size of testɑ=0.05,P<0.05)Results: 1、Expression of mi R-21 in AML increased compared with the control group(P <0.05), expression of mi R-21 in CR group was lower than the new diagnosed group and the refractory and relapse group(P <0.05), expression of mi R-21 in the refractory and relapse group is higher than the new diagnosed group, but no statistical significance between two groups(P >0.05).2、Expression of mi R-21 in different subtype of new diagnosed AML patients is different, expression in the M3 and M5 is higher than expression in the M1 and M2(P <0.05), without statistical significance between other subtypes(P >0.05).3、Expression of mi R-21 in primary treatment AML is positively correlated with the original naive in the bone marrow cells(P <0.05), and has no correlation with age, sex, white blood cells, hemoglobin concentration, platelet count and chromosome abnormality(P >0.05).4、Expression of PTEN m RNA in AML decreased compared with the control group(P <0.05), its expression in the new diagnosed group was lower than the CR group(P <0.05); expression of PTEN m RNA in the refractory and relapse group was lower than the CR group, but no statistical significance between two groups(P >0.05); without statistical significance between other groups(P >0.05).5、PTEN m RNA in different subtype of AML patients treated first expression is different, but no statistical significance(P >0.05).6、Expression of PTEN m RNA in primary treatment AML has no correlation with age, sex, the original naive in the bone marrow cells, white blood cells, hemoglobin concentration, platelet count and chromosome abnormality(P >0.05).7、Expression of mi R-21 and PTEN m RNA in AML has no correlation(P >0.05).Conclusion: 1、mi R-21 is excessive expression in patients with AML and its expression level is associated with disease states, FAB subtypes.2、mi R-21 expression level is positive with the percentage of bone marrow blast cells of AML.3、Expression of PTEN m RNA decrease in AML and its expression level is associated with disease states, but has no correlation with the FAB subtypes, clinical features of the patients.4、Aberrant expression of mi R-21 and PTEN m RNA in primary treatment AML show no correlation with chromosome abnormality.5、Expression of mi R-21 and PTEN m RNA in AML has no obvious correlation.
