| Objective:Tumor stroma, especially tumor-associated fibroblasts(CAFs), plays an important role in the incidence,growth,invasion,metastasis,prognosis and chemotherapy of breast cancer.In this study, explore the relationship among the protein expression of α-SMA,tumor characteristics of breast cancer and neoadjuvant chemotherapy before and after in the immunohistochemical methods, presume what the role of CAFs in tumor invasion and metastasis, and assess the efficacy of neoadjuvant chemotherapy in breast cancer,predict prognosis, and provide individualized treatment.Methods:This study recruited 56 female patients from 2010 to 2012 at the First Affiliated Hospital of Nanchang University,they are completed both in clinical and pathological data.another 10 normal breast tissues which distance to lesion above 5cm were confirmed by pathology. They were treated with 6-8 cycles of neoadjuvant chemotherapy containing anthracycline and / or taxane after obtained pathological data, After that,they had surgical treatment; By En Vision immunohistochemical method to detect α-SMA protein between breast cancer and normal breast tissue stroma fibroblast, as well as the expression of α-SMA protein in before and after neoadjuvant chemotherapy and tumor characteristics.Results:In breast cancer,the expression of α-SMA protein of CAFs is independent of the size of the tumor, P > 0.05, the difference was not statistically significant;However,in the group of the histological grade, clinical stage, axillary lymph node metastasis,recurrence and before and after neoadjuvant chemotherapy,the expression ofα-SMA protein was statistically significantin in breast invasive ductal carcinoma,P<0.05.Conclusion:1. The expression of α-SMA protein do not express in normal breast tissue fibroblasts, and high quality in breast cancer, especially in invasive breast cancer.2.α-SMA protein expression was significantly reduced in after neoadjuvant chemotherapy compared with before chemotherapy in breast cancer.3.α-SMA high expression in patients with high clinical stage, lymph node metastasis,probability of large chance of recurrence and distant metastasis after treatment, suggesting a poor prognosis. Thus,α-SMA may become a standard to judge malignant degree and prognosis. |
