Objective:To observe the effects of recombinant fusion protein IL-18 on inflammatory factors, such as IL-17 、 IFN-γ 、 IL-23 、 MIP and so on, in the Staphylococcus aureus infected mice, we performed an experiment to clarify the mechanism that the recombinant fusion protein IL-18 defend Staphylococcus aureus infection in vivo defense.Methods:SPF male BLAB / c mice were randomly divided into four groups(n = 10). Group A was the control group; Group B was Staphylococcus aureus infection group; Group C was protein of Staphylococcus aureus infections in the intervention group; group D was recombinant fusion protein group. The levels of IL-4, IL-17, IL-23, IFN- γ, TNF, G-CSF, Ig M in blood and BALF of mouse were observed through the ELISA method, the expression levels of MIP-1a and MIP-2β m RNA in lung tissue of mice were detected by real-time fluorescence quantitative PCR.Results:1. Serum IL-4 concentration levels in the groups B, C, and D are higher than that in group A(P < 0.05). BALF IL-4 concentration levels in the groups B, C, and D are higher than that in group A(P < 0.05). Among four groups, the IL-4 concentrations in BALF are higher than in serum.2. Serum IL-17 concentration levels in the groups B, and C are higher than that in group A(P < 0.05). The serum IL-17 concentration in group D is lower than that in group A. The BALF IL-17 concentration levels in the groups B, C, and D are higher than that in group A(P < 0.05). The concentration of IL-17 in BALF is higher than that in serum both group B and D.3. Serum IL-23 concentration levels in the groups B, C, and D are higher than that in group A(P < 0.05).BALF IL-23 concentration levels in Group B, C, and D are higher than that in group A(P < 0.05). The concentration of IL-23 in BALF is higher than that in serum both group A, B, and C. The concentration of IL-23 in serum is higher than that in BALF regarding group D.4. Serum IFN-γ concentration levels in the groups B, and C are lower than that in the group A(P < 0.05). The serum IFN-γ concentration in group D is higher than that in group A.(P < 0.05). BALF IFN-γ concentration levels in Group C and D are higher than that in group A(P < 0.05). The BALF IFN-γ concentration in group B is lower than that in group A.(P < 0.05). The concentration of IFN-γ in BALF is higher than that in serum both groups B, and C. The concentration of IFN-γ in serum is higher than that in BALF regarding groups A and D.5. Serum TNF concentration levels in groups B and C are higher than that in group A(P < 0.05). Serum TNF concentration levels in group D is no differences with group A(P = 0.239), the differences is no statistical significance. BALF TNF concentration levels in groups B, and C are higher than group A(P < 0.05). BALF TNF concentration levels in group D is no differences with group A(P = 0.488), the differences is no statistical significance. Among four groups, The TNF concentration is similar between BALF and serum.6. Serum G-CSF concentration levels in groups B, C, and D are significantly higher than that in group A(P < 0.05). BALF G-CSF concentration levels in Group B, C, and D are significantly higher than that in group A(P < 0.05). Among three groups(B, C, and D), the concentration of G-CSF in BALF is higher than that in serum.7. Serum Ig M concentration levels in group B, C, and D are Higher than that in group A(P < 0.05). BALF Ig M concentration levels in group B, C and D are Higher than that in group A(P < 0.05). Among four groups, the concentration of Ig M in BALF is significantly higher than that in serum.8. Group B, group C, group D MIP-1a and content in the lung tissue of mice are higher than group A(P < 0.05). The results are consistent with MIP-2β.Conclusion: The recombinant fusion protein IL-18 by mucosal immunity of mice, may increase the levels of IL-4, IL-17, IL-23, IFN-γ, TNF, G-CSF in mice serum and BALF after infected Staphylococcus aureus, improve the expression levels of MIP-1a and MIP-2β m RNA.
