| Objective To observe the correlation of the degree of damage in MMP-2 〠12 after chronic spinal cord jnjury by the experience in rabbits.And to observe the Dose-response relationship and therapeutic effect of different doses With Hong hua Injection after spinal cord injury in rabbits,with the purpose of providing experimental testimony to clinical treatment.Methods The models of chronic spinal cord injury in rabbits were induced with screwing the screw and the expression of matrix metalloproteinase-2ã€12 in chronic spinal cord injury was observed with ELASIA.The tissue sections were stained with immunohistochemistry and Real-Time PCR 〠RT-PCR. The Tarlov scales of hindlimb movement were carried out. The correlation about expression of MMP-2ã€12 with the degree of injury were analysed.Results1 Studies of the correlation of MMP-2ã€MMP-12 after spinal cord injury in rabbits(1)Before and after surgery the behavioral change of the rabbits was no statistical significance(P>0.05).(2)The MMP-2ã€12 were ecpressed throughout the process,and at the same time nodes A and B group,the difference was statistically significant(P<0.05);As the compression degree aggravating,MMP-2 and 12 express B group was obviously higher than that of group A, the difference is statistically significant(P<0.05).Group A were maintained at A relatively stable level at each time point.(3)The behavioral Point score lower than group A,group B with time differences statistically significant(P<0.05).2 Studies of the Dose-response relationship and therapeutic effect of Hong hua Injection after spinal cord injury in rabbits(1)Before surgery the behavioral change of the rabbits was no statistical significance(P>0.05).(2)After surgery the behavioral change of the rabbits was no statistical significance(P>0.05).(3)After the medication,The MMP-2 ã€12 were expressed reduce Compared with the preoperative in group Aã€Bã€C,it is statistically significant(P<0.05),Group D were Compared with the preoperative was no statistical significance(P>0.05),And after the medication Group B the MMP-2ã€12 were expressed reduce Compared with group Aã€Cã€D,it is statistically significant(P < 0.05).(4)After the medication,The behavioral change score high than thepreoperative in group Aã€Bã€C,it is statistically significant(P<0.05),Group B were high than group Aã€Cã€D,it is statistically significant(P<0.05).Conclusions The expression of MMP-2 and 12 obviously increased after chronic spinal cord injury in rabbit and with the degree of spinal cord compression,the expression of MMP-2and 12 also increased accordingly.By different doses of safflower injection after the intervention, the results show that the dosage of safflower injection on chronic spinal cord injury prevention and treatment of the rabbit curative effect is distinct, provides experiment basis for clinical medication. |
