Objective: Research on the inhibitory effect of luteolin on vitro proliferation of gastric cancer cell line MGC803, and its mechanism.Methods: 1.Luteolin evaluated by MTT assay and clone forming assay were put to MGC803 cells to inhibite their proliferation。2.The cell cycle changes were analyzed with the flow cytometry after staining propidium iodide(PI). Cell cycle-associated proteins were analyzed by Western blot.3.Using Annexin V-PI double staining to determine apoptosis ratio: in 0, 12.5, 25, 50 umol/L drug concentration effect on gastric cancer MGC803 cells 48 h, and DMSO control group, were detected.The Apoptosis-associated proteins were analyzed by Western blot.4.Autophagy-associated proteins were analyzed by Western blot.Results: 1.MTT assay:The IC50 for Luteolin against MGC-803 cells for 24 h,48h and 72 h are the 127.37umol/L,76.12umol/L,16.84umol/L.Cell colony formation inhibition assay demonstrated that cell clones were decreased with increasd concentration of Luteolin in MGC-8032. PI single staining flow cytometry found that cells were arrested in G2 phase, Western Blot results also proved that the cells in G2 phase after Luteolin effect on gastric cancer MGC80348 h.3.Flowcytometry Annexin V-FITC/PI double staining and Western blot experimental results show that: different concentrations of luteolin effect on gastric cancer MGC-803 cells, decreases as the concentration increased, and drug treatment cell early apoptosis rate and late apoptosis rate increased with the increase of drug concentration. Mcl-1, Bcl-xl, Caspase-3,Caspase-8 and Caspase-9 protein expression decreased with as the increase of drug concentration, and the amount of Bcl-2 protein changed little, the amount of Bax protein increased, thus causing such as Bcl-2/Bax, Mcl-1/Bax and Bcl-xl/Bax ratio decreased significantly. At the same time, can also see the Caspase-3 and PARP also appeared in the strip cutting. strip cutting can be seen from the Caspase-3 and PARP.4.Western Blot results indicated that, different concentrations ofluteolin effect on gastric cancer MGC-803 cells, the expression of P62 was decreased and Beclin-1and LC3 II was increased.Conclusion: 1.Luteolin blocked gastric cancer MGC803 cells at G2 phase, and put an effect on the cell growth by inducing autophagy.2. Luteolin induced apoptosis of gastric cancer MGC803 cells through mitochondrial pathway.
