AIM:To explore the expression of osmotic response element-binding protein(OREBP) at high glucose stress and its possible pathway.METHODS:(1)Human lens epithelial cells(HLECs) are cultured and divided into two groups:NC group, treated with normal concentration of D-glucose(25mmom/L), HG group, treated with high concentration of D-glucose(300mmom/L). The changing level of OREBP were detected by RT-PCR and western blot analysis.Pretreated HLECs with erk and P38 inhibitor fou 45 min and the activation of Erk/p38 MAPKs signaling pathway in HLECs were detected by Western blot analysis,.Then we deteched the level of OREBP by RT-PCR and western blot analysis.(2)A total of 103 patients(103eyes) suffered from DC and 154 patients(154 eyes) suffered from cataract that underwentpenetrating phaco+IOLat the Affiliated Hospital of Qingdao University from October 2013 to April 2014 composed experimental group and control group. 10 cases of healthy donor anterior lens capsule were used as blank controller. The expression of OREBP in lens subcapsular epithelial was detected by RT-PCR and immunofluorescence.(3)STZ induced diabetic rats were executed at 2w、4w、8w、10w、12w and The expression of OREBP in lens subcapsular epithelial was detected by RT-PCR.RESULTS:(1) OREBP was expressed in HLECs and elevated in 300mmol/L glucose condition.In high glucose stress, pretreated HLECs with U0126 and SB239063 for 45 min resulted in downregulation of OREBP.(2)OREBP expression was more higher in lens subcapsular epithelial in diabetic patient than healthy cataract patients and the two former were higher than healthy donors.(3)OREBP m RNA was elevated to the highest at 2 weeks and then gradually descend.CONCLUSION :(1)OREBP elevated in high glucose conditions.(2) OREBP expression was more higher in lens subcapsular epithelial in diabetic patient than healthy cataract patients and the two former were higher than healthy donors.(3)OREBP can be regulated by Erk and p38 MARK in HLECs.
