Objective:The expression of P- glycoprotein at different time points was detected by different concentrations of IL-17. To explore the regulation of IL-17 on P-gp Mediated Multidrug resistance. Methods:(1)RA was selected from 16 cases in the initial treatment group and 15 cases in the normal group. RA untreated group without any DMARDs and biological agents for treatment;(2)PBMC from the selected RA patients and The eripheral blood lymphocytes subsets count were detected by flowcytometry.(3)Collected blood samples of patients, Extract peripheral blood mononuclear cells, The percentage of positive cells of P-gp by flow cytometry, IL-17 with different concentrations to stimulate(10ng,100ng), The expression levels of P-gp in peripheral blood lymphocytes were measured at 0 hours, 24 hours and 48 hours respectively. Results:(1)The expression of P-gp in peripheral blood of patients with RA was elevated compared with the normal control group(P<0.05).(2)The expression of P-gp in RA patients was correlated with the Th% in the lymphocyte subsets(r=0.561, P<0.05).(3)The longer the IL-17 stimulus, the higher the expression of P-gp. At the same concentration of IL-17, the expression of P-gp increased at 48 hours. The difference was statistically significant between 0 hours and 24 hours(P < 0.05).(4)The concentration increase of IL-17, the expression of P-gp also increased. At the same time point, the higher the concentration of IL-17, the higher expression of P-gp. The difference was statistically significant(P < 0.05). Conclusions:(1)Compared with the normal control group, P-gp expression level of RA patients was increased.(2)In RA patients, There is a correlation between A the expression of P-gp and Th%.(3)The expression level of P-gp in patients with RA had a certain concentration and time dependence with IL-17.
