Objective:To improve the anti-carcinoma effect of lobaplatin(LBP),we can prepare the compound of carbon nanotubes and drugs(LBP-E2-PEG-MWCNT) and perform its characterization. We investigated the anti-tumor effect of LBP-E2-PEG-MWCNT on MCF-7 breast cancer in vitro and its influence in vivo. Method:1.Based on MWCNT, we loaded lobaplatin, estradiol and polyethylene and got the compound(LBP-E2-PEG-MWCNT). Finally we performed its characterization.2.By the MTT way, we detected respectively the inhibition ratio of lobaplatin, MWCNT-PEG, E2-PEG-MWCNT and lobaplatin-E2-PEG-MWCNT on MCF-7 cells.3.42 normal female mice were randomly divided into 7 groups, including MWCNT, PEG-MWCNT, E2-PEG-MWCNT, LBP-PEG-MWCNT, LBP-E2-PEG-MWCNT, lobaplatin and physiological saline control group. They were exposed by intravenous injection respectively at day 1 and day 7. The concentration of lobaplatin is 0.5mg/ml, 5mg/kg totally. We got blood from Eyeballs with EOTA anticoagulant and then tested blood routine test, and aspartate aminotransferase(AST), alanine aminotransferase(ALT), and creatinine(Cr).Killing the mice, we taken the tissue of heart, renal and liver,then respectively making pathological tissue examination. Result:1.MTT discovers the inhibition effect of human MCF-7 breast cancer cells: LBP-E2-PEG-MWCNT>LBP>MWCNT-PEG>E2-PEG-MWCNT. LBP-E2-PEGMWCNT is more potential inhibition to MCF-7 cells and has the time-dose relationship.2.The levels of mice platelet(PLT) decreased significantly in the groups of LBP-PEG-MWCNT,LBP-E2-PEG-MWCNT,LBP.(P<0.05) The platelet levels of LBP were significantly lower than LBP-PEG-MWCNT and LBP-E2-PEG-MWCNT.(P<0.05) The levels of serum aspartate aminotransferase(AST) were increased(P<0.05),while there is no significant difference in other groups. The levels of Hemoglobin(Hb), white blood cell(WBC),alanine aminotransferase(ALT) and creatinine(Cr) were no significant difference among groups. Conclusions:1. The preparation of compound(LBP-E2-PEG-MWCNT)is Feasible.2.Compared with lobaplatin, inhibition effect of LBP-E2-PEG-MWCNT produces stronger on human breast cancer cells(MCF-7) and has time-dependent effect. Compared with lobaplatin,the influence of LBP-E2-PEG-MWCNT is smaller in vivo.
