Objective:The research makes the Musca domestica larvae defensin recombinant protein’s inhibition effects of esophageal cancer cells as well as its possible mechanism explicit.Methods:1. Transform the successfully-constructed p GEX-6P-1 / MD expression vector into escherichia coli cells, induce the Musca domestica larvae defensin protein expression and purify the target protein.2. Put the different concentrations of Musca domestica larvae defensin recombinant protein in esophageal cancer cells and normal esophageal epithelial cells to detect the ability of cells’ s proliferation, invasion and migration.3. Detect the changes of biomechanics in biomechanical esophageal cells and normal esophageal epithelial cells before and after the effects of Musca domestica larvae defensin recombinant protein by using an atomic force microscope.Results:1. Musca domestica defensin recombinant protein can inhibit the proliferation of esophageal cancer cells KYSE150, but does not affect the normal esophageal epithelium SHEE.2. Musca domestica defensin recombinant protein can inhibit the invasion of KYSE150 esophageal cancer cells, but not affect the normal esophageal epithelium SHEE;3. Musca domestica defensin recombinant protein can inhibit the migration of the KYSE150 esophageal cancer cells, but has no effect in normal esophageal epithelium SHEE.4. By observing the morphological changes of cells before and after the effects of Musca domestica defensin recombinant protein with the help of AFM, we find that thesurface of esophageal cancer cells KYSE150 becomes rough, while the normal esophageal epithelium SHEE has no obvious change after the effects.5. The viscosity of esophageal cancer cells KYSE150 increases while the normal esophageal epithelium SHEE has no obvious change after the effects.6. The elasticity of esophageal cancer cells KYSE150 reduces meanwhile the normal esophageal epithelium SHEE has no obvious change after the effects.Conclusion:Musca domestica defensin recombinant protein has the ability to inhibit the proliferation, invasion and migration of the esophageal cancer cells by increasing tumor cell membrane surface viscosity, reducing the elasticity of membrane surface without affecting normal cells, which can be applied as an alternative drug of targeted-tumor therapy.
