Objective:To compare the expression of circulating tumour DNA(ct DNA) and carcino embryonic antigen(CEA) in patients with non-small cell lung cancer(NSCLC) before and after radical operation.To investigate the concentration of ct DNA can be a good molecular biological indicator for prognosis of NSCLC patients.Methods:We randomly assigned 5 NSCLC patients from the Department of Thoracic Surgery of Fujian Medical University Union Hospital,all patients underwent radical surgery.Collection the venous blood samples from the 5 NSCLC patients at three time points(3 days before the operation、1-3 days after operation、21-37 days after operation) for monitoring ct DNA levels,isolated plasma from veinal blood by using high velocity centrifugal,extracted plasma DNA utilize QIAamp Circulating Nucleic Acid kit,methods quantificationally measuring ct DNA level by Qubit quantitative instrument.Collection the venous blood samples from the 5 NSCLC patients at two time points(7 days before the operation and 21-37 days after operation) for monitoring CEA levels,quantitative measuring CEA concentrations by Roche E601 Full Automatic Immunological Analyzer.And then taking two related data to statistical analysis.Results : Each group of statistics are indicated with medians,The median concentration of ct DNA at the time of 3 days before the operation、1-3 days after operation、21-37 days after operation was 0.612、0.855、0.430ng/μL,respectively.3 days before the operation had higher concentrations of ct DNA than 21-37 days after operation,The concentrations of two groups showed significant difference(P<0.05).1-3 days after operation was highest concentrations of ct DNA than 21-37 days after operation and 3 days before the operation,but one samples of 3 days before the operation suggest pollution, and take it out,Compare the concentrations of ct DNA at the time of 3 days before the operation with 1-3 days after operation with no significant statistical difference(P>0.05).there were also no significant statistical difference between concentration of ct DNA at the time of 3 days before the operation and 21-37 days after operation(P>0.05).The median concentration of CEA at the time of 7 days before the operation and 21-37 days after operation was 3.90ng/m L、1.90ng/m L,,respectively,There has no statistically significant between them(P>0.05).Conclusion:Analysis of 5 patients with non-small cell lung cancer before and after radical surgery, the concentration of ct DNA is more sensitive for assessment of the change of NSCLC in tumour burden than CEA.The concentration of ct DNA can be a good bio-molecular markers for prognosis of NSCLC patients.And it is worth further study.
