Objective: The purpose of this study was to investigate the potential role of micro RNA-183-5p(mi R-183-5p) in the proliferation, invasion and metastasis of pancreatic cancer, and to identify the promising target genes of pancreatic cancer oncogenic mi R-183-5p.We based on q RT-PCR to investigate whether these oncogenic mi Rs may be useful as Pancreatic Carcinoma(Pa Ca)biomarkers.Methods: Initially, we used real-time PCR to detect the expression of mi R-183-5p and SOCS-6 in the pancreatic carcinoma cells, such as PANC-1,compare with HPDE6-C7 cells. To investigate the function of mi R-183-5p, we used a mi R-183-5p inhibitor in Cells proliferation, wound-healing, invasion and immigration assays. We forecasted the potential target genes using mi RDB, Pic Tar and Target SCAN. We used real-time PCR to detect and confirmed that the potential target gene is directly regulated by mi R-183-5p. We used Westernblot to detect the expression of SOCS-6 in pancreatic cancer tissue.Results: 1. The real-time q RT-PCR results: We identified the expression of mi R-183-5p in Pa Ca cells PANC-1 was up-regulated compared with HPDE6-C7 cell and the expression of SOCS-6 was down-regulate. Additionally, mi R-183-5p expression was correlated with SOCS-6. 2. Mi R-183-5p knockdown decreased cell growth and motility in Pancreatic Cancer cells and significantly increase the expression of SOCS-6. 3. SOCS-6 expression was also significantly lower in pancreatic carcinoma tissues compared with that in matched normal pancreatic tissues.Conclusion: Our data suggest that oncogenic mi R-183-5p was up-regulated in Pa Ca cells PANC-1 and may be useful as a pancreatic cancer biomarker and that inhibition of mi R-183-5p expression may be therapeutically beneficial as a pancreatic carcinoma treatment. The expression of SOCS-6 was down-regulate in Pa Ca cells PANC-1, it can inhibit the proliferation, metastasis and invasion ability of pancreatic cancer. We identified SOCS-6 as potential target gene of mi R-183-5p. The further study of mi R-183-5p-5p carcinogenic mechanism, is expected to make mi R-183-5p as a new target in the treatment of pancreatic cancer in the future.
