Object: To investigate maternal limb ischemic preconditioning on the change of PARP-1 and AIF expression in the fetal hippocampal neurons caused by distress-reoxygenation in the pregnant rats. looking for how to reduce brain damage of intrauterine distress,reducing its long-term sequelae to provide a theoretical basis and new idea in the clinical application. Methods: one male and three female SD(The Sprague Dawley) rats were put in a cage to make them mate.The pregnant SD rats were randomly received intraperitoneal anesthesia,paunched the rats and come out bicornuate uterus; Intrauterine distress model was constructed by blocking the arteriovenous of the uterine and ovarian with micro artery clips for 15 min following by loosen of the clips to recover reperfusion; at the same time,the model of LIP was established by 5min reperfusion in pregnant rats femoral artery blood flow in the right lower limb,the circulation was repeated three times. The pregnancy rats were stachastically assigned into Sham(S) group, Control(LIP) group, Fetal distress(FD) group and the LIP plus intrauterine distress(LIP+FD) group. Cesarean section occurred in pregnant rats(21 days gestation) to acquire live pups in each group after 2 days. The pups were decapitated, PARP-1 and AIF positive cells in the CA1 area of hippocampal neuron of fetal rat were observed by using light microscope in immunohistochemical staining. The expression of PARP-1 and AIF protein in hippocampal neuronal apoptosis of fetal rat was detected by Western blot test and expression changes of PARP-1 and AIF mRNA in hippocampal neuronal apoptosis of fetal rat were detected by the RT-PCR test. Result: 1)Compared with group S, The PARP-1/AIF positive cells in CA1 hippocampus, the expression changes of PARP-1 and AIF protein and PARP-1 and AIF mRNA in hippocampal neuronal apoptosis of fetal rat were significantly higher in FD group(P<0.05). 2) Compared with group S, The PARP-1 and AIF positive cells in CA1 hippocampus, the expression changes of PARP-1 and AIF mRNA and the PARP-1 and AIF protein in hippocampal tissue of fetal rat did not reach significant level in LIP group(P<0.05). 3) Compared with group S, The PARP-1 and AIF positive cells in CA1 hippocampus, the expression changes of the PARP-1 and AIF mRNA and the PARP-1 and AIF protein in hippocampal tissue of fetal rat were apparently higher in group LIP+FD(P<0.05). 4) Compared with group FD, The PARP-1 and AIF positive cells in CA1 hippocampus, the expression changes of protein PARP-1 and AIF and the protein PARP-1 and AIF mRNA in hippocampal tissue of fetal rat were apparently lower in LIP+FD group(P<0.05). Conclusions: PARP-1 and AIF in hippocampal neuronal apoptosis of fetal rat induced by distress-reoxygenation in rats was significantly downregulated by maternal limb ischemic preconditioning, thereby LIP can inhibit fetal hippocampal neurons PARP-1 dependent programmed cell death(Parthrantos)induced by intrauterine distress-reoxygenation in the pregnant rats.
