Objective:To study the therapeutic effect of allitridum on immunological hepatic fibrosis model rats which induced by porcine serum, provide an experimental basis for the treatment of liver fibrosis allitridum. We will discuss it from 4 aspacts: 1. The therapeutic effect of allitridum on immunological hepatic fibrosis model rats which induced by Porcine serum. 2. The preventive effect of allitridum on immunological hepatic fibrosis model rats which induced by Porcine serum. 3. The effect of allitridum on extracellular matrix. 4.The regulation effect of allitridum on inflammatory factor and liver immune fuction.Methods:Models of hepatic fibrosis were induced by Porcine serum in Wistar rates for 12 weeks. 25 male and 25 female SD rats were randomly divided into 5 groups as follow: blank control group, model group, low concentration prevention group, middle concentration prevention group, middle concentration treatment group. Model group was duplicated intraperitoneal injection of porcine serum biweekly at dose of0.5ml/time for 12 weeks. Low concentration prevention group, middle concentration prevention group was treat with allitridum for 12 weeks with the dosages of 11, 22 mg/(kg·d). Middle concentration treatment group was treat with allitridum from 8 week with the dosages of 22 mg/(kg·d). At the end of 12 weeks,animals were sacrificed to obtain blood sample and liver tissue samples. Test items include:(1) The hepatic histological changes were also observed by optical microscopy after HE and picrosirius red staining.(2)The expression level of ALT, AST, TP, ALB, TB, DB, IB was detected with Full-automatic Enzyme.(3)The expression level of CⅠ, PⅢP, CⅣ was detected with method of ELISA.(4) The expression level of type Ⅰand Ⅳcollagen protein was detected with method of Western blot.(5) The expression level ofα-SMA was detected with method of ELISA.(6) The expression level of TGF-β1, IL-10, IL-6, IFN-γ,TNF-α was detected with method of ELISA.Results:(1) Pathohistological observation: The pathobiology results showed that allitridum could attenuate the degree of hepatic fibrosis. Compared with the model group, the degree of fibrosis was milder in the low concentration prevention group, middle concentration prevention group, middle concentration treatment group.(2) The level of liver fuction: The expression level of ALT, AST, TP, ALB, TB, DB, IB was within normal range. There were no significant differences between the 5 groups, the difference was not statistically significant(P> 0.05).(3) The level of ECM : Compared with the model group, the expression level of CⅠ 、 PⅢNP was significantly lower(P <0.01) in low concentration prevention group, middle concentration prevention group,middle concentration treatment group. Compared with the model group, the expression level of C IV was significantly lower(P <0.01) in low concentration prevention group, middle concentration prevention group,middle concentration treatment group.(4) The level of ECM: Compared with the model group, type Ⅰand Ⅳcollagen protein expression was significantly lower(P <0.05) in low concentration prevention group, middle concentration prevention group,middle concentration treatment group.(5) The level of α-SMA: Compared with the model group, the expression level of α-SMA was significantly lower(P <0.01) in low concentration prevention group, middle concentration prevention group, middle concentration treatment group.(6)The level of inflammatory factor: Compared with the model group, the expression level of IL-6, TNF-αwas significantly lower(P <0.05) in low concentration prevention group, middle concentration prevention group. Compared with the model group, the expression level of TGF-β1 was significantly lower(P <0.05)in low concentration prevention group, middle concentration prevention group, middle concentration treatment group. Compared with the model group, the expression level of IFN-γ was significantly higher(P<0.05) in low concentration prevention group, middle concentration prevention group, middle concentration treatment group. Compared with the model group, the expression level of IL-10 was no significant differences between the 5 groups, and the difference was not statistically significant(P> 0.05).Conclusions:1. Allitridum has protective infection on hepatic fibrosis, and the preventive effect had an advantage over therapeutic effect.2. Allitridum can protect liver cells, and has no impairment on liver function.3. Allitridum can edreasede the expression of ECM and inhibt proliferation of HSC.4. Allitridum can regulate inflammatory factor and keep the banlance of Th1/ Th2 cytokines.
