Background:Non-alcoholic fatty liver disease (NAFLD) is a kind of chronic liver diseases caused by liver damage dimensions apart from alcoholic and other specific factors, the main feature is hepatic steatosis, has gradually become the common chronic liver disease in China and western countries. Secreted frizzled-related protein 5(SFRP5)was a kind of endogenous anti-inflammatory fat factor which can inhibit Wnt signaling pathways. Recent years the expression and significance of SFRP5 was studied in many metabolic diseases, such as type 2 diabetes, obesity and coronary heart disease. More and more studies has shown that NAFLD has a close relation with obesity, type 2 diabetes, insulin resistance, hyperlipemia, cardiovascular disease, however, there is no research about the expression of SFRP5 in NAFLD and its correlation with serum lipids and other relative factors in China and other countries.Objective:To investigate the levels of serum SFRP5 in NAFLD and the correlation of SFRP5 with serum lipids and other relative factors.Methods:From Oct.2013 to Jul.2014,58 subjects who were diagnosed with NAFLD from the second affiliated hospital of Chongqing medical university were considered as the experimental group, and other 58 healthy subjects who were selected from Center of Health of the second affiliated hospital of Chongqing medical university were served as normal controls(NC), On account of the abdominal ultrasonography, the liver fatty change were divided into mild, medium and heavy by two experienced ultrasound doctors, according to the degree of steatosis NAFLD group will be divided into three subgroups. All research objects will detect the level of Fasting Plasma Glucose(FPG),HBAlc,Total Cholesterol(TC), Triglyceride (TG),High-density lipoprotein(HDL), Low density lipoprotein(LDL), Free fatty acid(FFA), Apolipoprotein B(ApoB), Uric acid, Alanine aminotransf-erase(ALT),Aspartate aminotransferase(AST),Alkaline phosphatase (ALP) Gamma-glutamyl transpeptidase(y-GGT), Total bilirubin (TB),Conjugated bilirubin(CB),Unconjugated bilirubin(UCB), Prothrombin activity(PTA) in the clinical laboratory of the second affiliated hospital of Chongqing medical university, ELISA was used to detect the SFRP5 level, then analyze the correlation of SFRP5 with other relative factors.Results:The levels of BMI、SBP、DBP、HbAlc、TG、TC、LDL-C、 ApoB、Uric acid、ALT、AST、y-GGT、SFRP5 were significantly higher in NAFLD group than in NC group (P<0.05 或 P<0.01), Pearson correlation analysis showed that serum SFRP5 was negatively associated with TC、HDL-C、Apo-B (P<0.05 or P<0.01); Spearman correlation analysis showed that the serum SFRP5 level was positively associated with the liver fatty content (P=0.029, r=0.287) Multivariate linear regression analysis showed that TC was SFRP5’s independent relative factor.Conclusion:Serum SFRP5 level was increased in NAFLD subjects and was negatively associated with TC and positively associated with the liver fatty content, it suggested that in order to counteract liver fat accumulation, metabolic stress and insulin resistance the SFRP5 exist chronic or acute compensatory mechanism, it may contribute to delay the development of nonalcoholic fatty liver disease.
