Objective:To observe the effects of chuanxiong drug serum on expression of Toll-like receptor 4(TLR4) and its down stream signaling components myeloid differentiation factor 88(MyD88) in rat hepatic stellate cells.Methods:SD rats were randomly divided into 5 groups:Normal control group (n=10), colchicine group (n=10), the low concentration chuanxiong drug serum group (n=10), middle concentration chuanxiong drug serum group (n=10), and high concentration Chuanxiong drug serum group (n=10). The Blood was collected after gastric perfusion 2 hours at 7th day. HSCs were cultured in vitro and stimulated with LPS, then were treated separately with the serum of normal saline, colchicine,Chuanxiong compound(low, medium and high concentration) for 24 hours. The expression of TLR4, MyD88 mRNA and protein were detected by RT-PCR and Western blot.Results:LPS could enhance the expression of TLR4, MyD88 mRNA and protein compared with normal control group (P<0.01). Compared with model group, the expression of TLR4, MyD88 mRNA and protein were decreased significantly in Chuanxiong compound group (P< 0.05, P<0. 01)Conclusion:Chuanxiong drug serum inhibits the expression of TLR4, MyD88 mRNA and protein, which may play an important role in Toll-like receptor 4 signaling pathways in hepatic fibrosis. The Chinese medicine chuanxiong inhibit hepatic fibrosis in a dose dependent manner.
