Objective:In order to study the effects of 1-tetradecyl-3-methyl-imidazolium salt ([Ci4mim]BF4) on treated mice immune system and its possible molecular mechanism, we design the in vivo and in vitro tests.Methods:1) Three different dosage groups (1.4 mg/kg、2.8 mg/kg、5.6 mg/kg) and one blank control group were set up. After KM mice were treated with [C14mim]BF4 solution by intragastric administration for 21 days, the effects of [Ci4mim]BF4 on organ coefficient, serum biochemical index and histomorphology were observed.2) Test the effects of ionic liquids on the humoral immunity by plaque forming cell assay.3) Using flow cytometric analysis to determine the changes of T cell subsets of mouse spleen and NK cell ratio.4) In vitro, T cell proliferation and NK cell activity were detected by MTT assay.Results:Compared with the control group, absolute number of WBC and granulocyte, immunoglobulin G and M were reduced; the liver coefficient and spleen coefficient increased obviously, and thymus coefficient significantly reduced. The phenomena of thymic atrophy, spleen cells edema, hepatocytes hyperemia and necrosis can be observed by conducting pathological tests. Reduced number of antibody was generated in hemolytic plaque test; lymphocyte stimulation index and activity of NK cells were decreased, while the ratio of CD3+CD4+ and CD3+CD8+ in spleen lymphocytes were increased.Conclusion:After exposure to [Ci4mim]BF4, the lumphocytes, immune globulin G and M in blood of KM mice were reduced, the normal structure of their immune organs were destroyed, such as thymus atrophy, spleen and liver congestion enlargement, while B lymphocyte secretion activity, T lymphocyte proliferation and NK cell killing activity were inhibited. These results indicated that [C14mim]BF4 has a certain immunotoxicity which could damage immune organs of mice, and inhibit its specific immune and non-specific immune function.
