| Purpose To observe the effect of Qinghuagushenpaidu decoction on chronic renal failure through the activate of transcription factor NF-κB, which promotes the expression of inflammatory molecule---- MCP1 and investigate the expression of NOX4, which affected by Qinghuagushenpaidu decoction.Methods Marked 108 male SPF Sprague Dawley rats by weight, then divided into sham-operated group (SOR group, eighteen rats) and UUO group (ninety rats). UUO group was taken with left ureteral occlusion, while the SOR group was operated with left ureter separation. UUO group was divided into 5 groups randomly after molded, eighteen rats in each group:western medicine group, high solubility of decoction, medium solubility of decoction, low solubility of decoction, control group, administrated all the rats by gavage at the 2nd day after molding. Executed six rats in each group at the 7th,14th,21st day, abrosia only for 24 hours and collected 24h urine before executed, measured 24h urine protein. Blood samples were collected from heart, then measured serum creaeinine (Scr), blood urea nitrogen (BUN), Uric acid (UA). Observed the pathomorphism change of left kidney, and detected the expression of MCP-1ã€NF-κBã€NOX4 by immunohistochemical technique(IHC).Results 1. For 24h urine protein:there is no significant difference(P>0.05) between all the group in terms of 24h urine protein at the same time.2. For Scr, BUN:At the same time, the BNU, Scr were significant higher in control group than SOR group(P<0.05) and the other four UUO groups; The differences of the two indexes between western medicine group, high, medium, low solubility of decoction groups were not apparent(P>0.05).3. The pathomorphism change of left kidney by HE stain:The anatomy of kidney tubules and the staining of nephridial tissues were normal in SOR group. At the 7th day, there was apparent inflammatory cell infiltration in control group, renal cortexe became thinner over time, then could see renal interstitial fibrosis, but the glomeruli was normal. At the same time, compared with control group, the inflammatory cell infiltration, thin renal cortexe were slighter in the other four UUO groups, could see renal interstitial fibrosis, either. There was no significant difference between the other fours UUO groups.4. the expression of MCP-1ã€NF-κBã€NOX4 by immunohistochemical technique(IHC):(1) The expression of NF-κB in SOR group was slight, which was significant higher in control group at the same time(P<0.01); Compared with the other four UUO groups, the expression of NF-κB was higher in control group(P<0.05). There was no apparent difference between different solubility of decoction groups(P>0.05).(2) The expression of MCP-1 in SOR group was thimbleful, which was apparent higher in control group at the same time(P<0.01); And it was significant lower in the other four UUO groups than in control group(P<0.05), There was no apparent difference of The expression of MCP-1 between different solubility of decoction groups(P>0.05).(3) The expression of NOX4 in SOR group was slight, which was apparent higher in control group at the same time(P<0.01); And it was significant lower in the other four UUO groups than in control group(P<0.05), There was no apparent difference of The expression of NOX4 between different solubility of decoction groups(P>0.05).Conclusion:1. Qinghuagushenpaidu decoction could protect kidney for it can reduce Scr, BUN of UUO rates.2. Qinghuagushenpaidu decoction could change the pathomorphism of kidney, delay the development of renal interstitial fibrosis, alleviate the damage of kidney in UUO rates.3. The renal interstitial fibrosis process may be intervened by Qinghuagushenpaidu decoction, for ti could reduce the expression of NF-κBã€MCP-1ã€NOX4.4. Qinghuagushenpaidu decoction may interdict the signal transduction pathway of NF-κB→+NF-κB, then delay the development of renal interstitial fibrosis. |
