Objective: The incidence of colorectal cancer is a multifactorial,complex multi-step evolution, gene expression abnormality inwhich plays an important role.Multicenter study shows that microRNA expression changes close to the clinical pathology of colorectal cancer are important factors leading to progress and invasion and metastasis of colorectal cancer.Now several mi RNAs expression abnormality in colorectal cancer, which microRNA-92 a is a member of a large number of miRNAs,and numerous studies have shown that microRNA-92 a is closely related to the occurrence and development of colorectal cancer.Therefore, further explore the pathogenesis of colorectal cancer, and find colorectal cancer early diagnosis and prognostic molecular markers for clinical colorectal metastasis of early detection and prompt treatment is important.This test is to verify that the miR-92 a in colorectal cancer development and adjustment mechanism in the invasion and metastasis.Methods: Prediction miR-92 a of target gene through bioinformatics software, then use dual luciferase reporter system for validation.In addition,using qRT-PCR, Western blot analysis and immunohistochemical detection of miR-92 a and target gene expression by MTT method, cell migration and invasion experiment and detection miR-92 a miRNA transfection on cell proliferation, migration, and invasion impacts.Results: 1.miR-92 a expression is significantly higher in patients with Colorectal carcinoma and colorectal cancer with lymph node metastasis significantly higher expression in patientswithout metastasis.(P<0.05)2.miR-92 a of colorectal cancer cell line SW480, HT29, andHCT116 expression higher than that in normal colorectal epithelial cells FHC.3.Ectopic expression of miR-92 a to promote cell proliferation, migration and invasion.4.In experimental determination of Xenografts in nude mice and miR-92 a can promote colorectal cancer cell proliferation and tumour formation.5.High-level expression of mi R-92 a induces Epithelial-mesenchymal cell transformation(EMT).6.MiR-92 a overexpression inhibits the expression of PTEN and negatively related to both.Conclusion: High expression of miR-92 a can promote the occurrence of colorectal cancer.High expression of miR-92 a induces Epithelialmesenchymal transformation(EMT), decreased adhesion between the cells and morphological changes, which facilitate the transfer of cancer cells.miR-92 a by regulating the expression of PTEN, which regulates cell proliferation in Colorectal, migration and invasion.Therefore,mi R-92 a could become a colorectal cancer diagnosis and treatment of a new target.
